摘要 目的探讨Gli抑制药GANT61对卵巢癌细胞生长和转移抑制作用的研究。方法选取40只雌性昆明大鼠,依据随机数表法按1∶1分为GANT61组、对照组,每组20只,所有大鼠均建立卵巢癌模型,对照组注射1∶4的无水乙醇:玉米油混合液,GANT61组皮下注射10 mg/ml的Gli抑制药GANT61,Western blot(WB)法检测Gli1、Gli2、Ki67、血管内皮生长因子(vascular endothelial growth factor,VEGF)表达水平,克隆形成实验检测卵巢癌细胞增殖能力,Transwell侵袭实验检测卵巢癌细胞转移能力。结果 GANT61组肿瘤质量和第7、15天肿瘤体积显著低于对照组,差异有统计学意义(P<0.001);GANT61组Gli1、Gli2、Ki67、VEGF的WB值显著低于对照组,差异有统计学意义(P<0.001);GANT61组细胞存活克隆计数及穿膜细胞数显著低于对照组,GANT61组死亡细胞计数显著高于对照组,差异有统计学意义(P<0.001)。结论 Gli抑制药GANT61可能通过下调Gli1、Gli2、Ki67、VEGF蛋白表达而抑制卵巢癌细胞的生长和转移。 Objective To discuss the role of Gli inhibitor GANT61 in inhibiting cell growth and metastasis in ovarian cancer.Methods Forty female Kunming rats were selected using the random number table.These rats were divided into GANT61 group and control group,with 20 in each.An ovarian cancer model was established in each of these rats.The control group was subcutaneously injected with 1:4 mixture of absolute ethyl alcohol and corn oil,while the GANT61 group was given subcutaneous injection of 10 mg/ml GANT61 Gli.The expression levels of Gli1,Gli2,Ki67 and VEGF were detected by Western blot(WB),the proliferation of ovarian cancer cells was detected by cloning experiments,and the migration of ovarian cancer cells was detected by Transwell invasion experiments.Results The tumor mass and tumor volume at 7 d and 15 d of the GANT61 group were significantly smaller than those of control group,and the difference was statistically significant(P<0.001).The WB values of Gli1,Gli2,Ki67 and VEGF in the GANT61 group were significantly lower than those of the control group,and the difference was statistically significant(P<0.001).The number of surviving cell clones and the pleural cell number of the GANT61 group were significantly smaller than those of the control group,but number of dead cells was significantly larger,and the difference was statistically significant(P<0.001).Conclusion Gli inhibitor GANT61 may inhibit the growth and metastasis of ovarian cancer cells by reducing the expressions of Gli1,Gli2,Ki67 and VEGF protein.
