摘要
目的观察不同糖尿病病程患者胫后神经中蛋白激酶C-β_2(protein kinaseβ_2,PKC-β_2)、S-100蛋白的表达情况,为糖尿病周围神经病变与PKC-β_2、S-100蛋白关系的研究提供依据。方法选取2012年5月至2017年6月在河北北方学院附属第一医院骨外科因糖尿病足行高位截肢患者截下的肢体共50例,均于大腿中下1/3截肢,手术时留取截肢肢体胫后神经,应用免疫组织化学方法观察不同糖尿病病程患者胫后神经PKC-β_2及S-100蛋白表达的差异。结果免疫组织化学染色结果显示,不同糖尿病病程患者的糖尿病足胫后神经均有PKC-β_2、S-100蛋白表达,且随糖尿病病程延长,糖尿病足胫后神经中PKC-β_2表达水平逐渐增高,S-100蛋白表达水平逐渐降低,差异均有显著性(P<0.05)。结论 PKC-β_2表达增多、S-100蛋白表达减少可能通过某种机制参与了糖尿病周围神经病变的发生发展。
Objective To observe the expression of protein kinase C-β2(PKC-β2)and S-100 protein in the posterior tibial nerve in patients with different pathologies,and to provide a certain theoretical basis for the relationship between diabetic peripheral neuropathy and PKC-β2,S-100 proteins.Method Fifty cases of amputated limbs of patients with high amputation due to diabetic foot(DF)performed at the first affiliated hospital of Hebei Northern College from May 2012 to June 2017 were taken.1/3 amputation was performed in the middle and lower thighs.The posterior tibial nerve of the amputated limb was retained during surgery.For the remaining specimens,we used immunohistochemical methods to observe the differences in posterior tibial neuropathy in different pathologies:PKC-β2 and S-100 staining,Variations in the expression of PKC-β2 and S-100 proteins in the posterior tibia nerves with different pathologies were measured under microscope.Result The immunohistochemical results showed that different course of diabetic foot posterior tibial nerve were S-100 protein,PKC-β2 expression of different course of diabetes foot posterior tibial nerve expressed significant difference,statistically significant(P<0.05).Conclusion Increased expression of PKC-β2 and decreased expression of protein S-100 may be involved in the occurrence and development of diabetic peripheral neuropathy through some mechanism.
