封丘金银花炭疽病病原的分离和鉴定 被引量:4

摘要 为明确河南封丘金银花炭疽病病原种类,对采自封丘的金银花炭疽病病叶,依据柯赫氏法则进行分离、纯化、回接和再分离,证明获得的分离菌株是金银花炭疽病的致病菌.对其培养性状、在寄主上的形态特征及萌发方式进行观察,并对其r DNA-ITS序列进行分析,结果表明:致病菌株在PDA培养基上菌落呈白色,后期菌落中央颜色变暗,生分生孢子盘;分生孢子盘具黑色刚毛,分生孢子长圆柱形,两端钝圆,单细胞.核糖体DNA-ITS序列与GenBank数据库胶孢炭疽菌同源性达99%.将封丘金银花炭疽病病原菌鉴定为胶孢炭疽菌Colletotrichum gloeosporioides. In order to determine the pathogen of Lonicera japonica anthracnose in Henan Province,infected leaves were collected from Fengqiu,Henan Province.The pathogen was isolated,back-inoculated,re-isolated by Koch’s postulates and was identified according to morphological and culture characteristics and the sequence of ribosomal DNA-ITS.The results showed that the fungus colonies on PDA were white,later pale yellow.Acervuli with setae formed in the center of the colonies.Conidia long cylindrical,obtuse at both ends,single-celled.rDNA-ITS sequence analysis indicated that the pathogen and Colletotrichum gloeosporioidesisolate COL4 were 99%in homology.he pathogen was identified as Colletotrichum gloeosporioides.
