HLA-DP基因rs3077和rs9277535位点多态性与汉族人群肺癌的关联性研究

摘要 目的探讨汉族人群中白细胞抗原(HLA)-DP基因多态性与肺癌遗传易感性的关联性。方法应用Taq?Man-MGB探针方法检测rs3077和rs9277535位点基因型,比较新发肺癌组患者和对照组不同基因型与肺癌患病风险的关系。结果 rs3077和rs9277535位点突变纯合基因型AA明显增加肺癌的发病风险(OR分别=1.84、1.91,P均<0.05);单倍型分析显示,A-A单倍型携带者肺癌发病风险更高(OR=1.40,P<0.05);对比未携带危险等位基因,携带危险等位基因发生肺癌的风险明显升高,并且肺癌发生风险随携带基因个数增加呈现明显的剂量反应关系(P趋势=1.18×10-11)。结论 HLA-DP基因rs3077和rs9277535两个位点会增加肺癌的发病率,HLA-DP基因多态性与中国汉族人群肺癌的发病风险相关。 Objective To investigate the association between polymorphism of human leukocyte antigen(HLA)DP gene and genetic susceptibility of lung cancer.Methods The genotypes of rs3077 and rs9277535 were detected by Taq-Man-MGB probe,and the relationship between different genotypes and the risk of lung cancer in new lung cancer patients and healthy controls was compared.Results The homozygous genotype AA of rs3077 and rs9277535 loci signifiCantly increased the risk of lung cancer(OR=1.84,1.91,P<0.05).The haplotype analysis showed that carriers of A-A haplotype had higher risk of lung cancer(OR=1.40,P<0.05).Compared with carriers of zero risk alleles,the risk of lung cancer in carriers of risk alleles was increased significantly,and with the increasing of the number of risk alleles,the risk of lung cancer showed a significant dose-response relationship(P trend=1.18×10^-11).Conclusion HLA-DP gene rs3077 and rs9277535 loci can significantly increase the incidence of lung cancer.HLA-DP gene polymorphism is associated with the risk of lung cancer in Chinese Han population.
