摘要
肿瘤干细胞(cancer stem cells,CSCs)假说提出了关于肿瘤发生的新解释,认为肿瘤组织主要存在两种类型的肿瘤细胞,即普通的肿瘤细胞和极少量肿瘤干细胞,这些普通的肿瘤细胞构成肿瘤的主要成分,且具有组织来源特异性;而对于那些含量极少的肿瘤干细胞,在启动肿瘤的发生、发展中起着至关重要的作用,具有极强的致瘤能力和自我更新并多向分化的潜能。研究发现,CSCs与肿瘤的复发、远处转移、不良预后及多重耐药性等密切相关,因而成为肿瘤的根治难题。CSCs的鉴定和分离是肿瘤干细胞研究进展的颈瓶,对制定有效的治疗策略有着重要意义,利用流式细胞术(flow cytometry,FCM)对肿瘤干细胞进行识别和分选,使对肿瘤干细胞功能特征的进一步研究成为可能。本文就流式细胞术在肿瘤干细胞中的鉴定和分选等应用进展作一简要综述。
The cancer stem cell (CSCs) hypothesis proposes a new interpretation of tumorigenesis.It is believed that there are mainly two types of tumor cells in tumor tissues,namely common tumor cells and a very small number of tumor stem cells.Common tumor cells constitute the main component of tumors and have tissue-derived specificity, the tiny amount of tumor stem cells play a crucial role in initiating the occurrence and development of tumors,and have strong tumorigenic ability,potential of self-renewal and multi-directional differentiation.Studies show that CSCs are closely related to tumor recurrence,distant metastasis,poor prognosis,and multi-drug resistance,thus becoming a challenge for radical treatment in tumor.The identification and isolation of CSCs is the "neck of bottle" of cancer stem cell research and is of great significance for the development of effective therapeutic strategies for cancer.The application of flow cytometry (FCM) to identify and sort cancer stem cells make it possible to further analyze the functional features of these CSCs.This article gives a brief review of the application of flow cytometry in the identification and sorting of cancer stem cells.
