摘要 目的通过对老龄系统性红斑狼疮小鼠(TC)皮肤症状的观察和鉴定,探讨其是否可以作为一种研究皮肤型狼疮的小鼠模型。方法观察TC小鼠出现皮肤症状的年龄,对比同龄小鼠与C57BL/6小鼠的皮肤症状,ELISA法检测小鼠血清抗dsDNA抗体及抗ANA抗体,HE染色检测小鼠皮肤病理学症状。结果 2/3的TC小鼠出现毛发脱失、皮肤溃烂等症状,其病变最早发生于40周龄,皮肤病变的狼疮小鼠血清抗dsDNA抗体和抗ANA抗体滴度明显高于C57BL/6小鼠(P<0.05);HE染色结果显示狼疮小鼠角质层缺失,上皮层连续性中断,真皮层大量淋巴细胞浸润。结论狼疮小鼠在40周后会出现皮肤病变,显示该小鼠可作为一种研究慢性皮肤型狼疮的疾病模型。 Objective To observe and identify the skin alterations of aged TC mice with systemic lupus erythematosus (SLE), and to explore if they can be used as an animal model for studying cutaneous lupus erythematosus. Methods To determine the age of TC mice when skin changes start to occur, and to compare the skin alterations between C57BL/6 and TC mice at the same age. Serum levels of anti-double-stranded (ds) DNA antibodies and anti-nuclear antibodies (ANAs) were detected by ELISA. Pathological changes in the mouse skin were detected by hematoxylin and eosin (HE) staining. Results Two-thirds of TC mice developed hair loss and skin ulceration. The lesions first occurred at 40 weeks of age. Serum titers of anti-dsDNA and anti-ANA antibodies in TC mice with skin lesions were significantly higher than those of C57BL/6 mice ( P < 0.05).Pathological examination observed loss of stratum corneum, interruption in the epithelial layer, and extensive lymphocyte infiltration in the dermid of SLE mice. Conclusion Skin lesions occur in SLE-prone TC mice after 40 weeks of age, suggesting that aged TC mice may serve as a useful animal model for studies of chronic cutaneous lupus erythematosus.
