奥美拉唑对大鼠反流性食管黏膜损伤后氧化应激及细胞增殖相关蛋白表达的影响 被引量:8

Effect of omeprazole on oxidative stress and expression of cell proliferation-related protein in reflux esophageal mucosa injury rats

摘要 目的观察奥美拉唑对大鼠反流性食管黏膜损伤后氧化应激及细胞增殖相关蛋白表达的影响。方法用食管下段与胃底吻合术构建反流性食管黏膜损伤大鼠模型,并随机分为模型组及低、中、高剂量实验组,各10只,另选10只作为假手术组。低、中、高剂量实验组大鼠于术后第6天灌胃给予奥美拉唑10,20,40 mg,假手术组及模型组则灌胃等体积0. 9%Na Cl,每日1次,连续干预8周。以苏木精-伊红(HE)染色观察大鼠食管黏膜病理变化;以生化试剂盒法及免疫组化法分别检测大鼠食管黏膜组织氧化应激指标及增殖细胞核抗原(PCNA)表达情况。结果假手术组食管黏膜光滑、结构完整、黏膜基底膜清晰可见,厚度适中且细胞排列整齐,黏膜组织无充血、水肿,亦无炎症或白斑表现;模型组食管黏膜增厚,基底层细胞数量增多,角化层增厚,局部黏膜组织呈乳头样突起,伴有点状或片状充血,部分可见糜烂并伴有大量炎性细胞浸润,少量树皮样增生白斑;低、中、高剂量实验组病理变化较模型组逐渐减轻。假手术组、模型组及低、中、高剂量实验组大鼠食管黏膜组织丙二醛(MDA)含量分别为(4. 75±1. 03),(10. 69±1. 15),(8. 37±1. 12),(7. 01±1. 06),(5. 84±0. 97)nmol·mg^(-1)pro;超氧化物歧化酶(SOD)含量分别为(168. 97±10. 21),(93. 26±8. 42),(112. 63±8. 15),(129. 07±8. 01),(159. 33±7. 65) U·mg-1pro; PCNA蛋白平均灰度值分别为128. 65±8. 21,29. 33±4. 26,46. 81±7. 36,75. 47±8. 01,109. 34±8. 19,差异均有统计学意义(均P <0. 05)。结论奥美拉唑可抑制反流性食管黏膜损伤大鼠MDA表达,促进SOD表达,上调PCNA基因表达,改善其黏膜损伤,修复食管黏膜上皮。 Objective To explore the effect of omeprazole on oxidative stress and expression of cell proliferation-related protein in reflux esophageal mucosa injury rats.Methods Forty healthy male rats were given lower esophagus and stomach bottom anastomosis to induce reflux esophageal mucosa injury and divided randomly into model group and experimental-L,-M,-H groups,10 rats in each group,while 10 rats were assigned to sham-operation group.Experimental-L,-M,-H groups were gavaged with 10,20,40 mg omeprazole at the 6 thd after operation,and sham-operation group and model group were gavaged with 0.9%NaCl,once daily, 8 weeks in total.The pathological changes of esophageal mucosa in rats were observed by hematoxylin and eosin( HE)staining;the oxidative stress indexes and the expression of proliferating cell nuclear antigen( PCNA) were detected by biochemical kit method and immunohistochemical method.Results Esophageal mucosa in sham-operation group was smooth,structure was complete and mucosa basement membrane was clearly visible,thickness is moderate and cells were ordered,no edema and inflammation or white spot in mucosal hyperemia;Esophageal mucosa in model group were thickened,basal layer cells were increased,keratin layer were thickened;Local mucosal tissue showed papillary projection,with dot or flake hyperemia and part of the visible erosion and accompanied by a large number of inflammatory cells infiltration,a small amount of bark hyperplasia white spot now;the pathological changes of esophageal mucosa tissue in experimental-L,-M,-H groups relieved gradually compared with model group.The malondialdehyde( MDA) content in esophageal mucosa of rats in experimental-L,-M,-H groups were( 4.75±1.03),( 10.69±1.15),( 8.37±1.12),( 7.01±1.06),( 5.84±0.97) nmol·mg^-1 pro,respectively;The superoxide dismutase( SOD) content were( 168.97±10.21),( 93.26±8.42),( 112.63±8.15),( 129.07±8.01),( 159.33±7.65)U·mg^-1 pro,respectively;The PCNA protein average grey value were( 128.65±8.21),( 29.33±4.26),( 46.81±7.36),( 75.47±8
