摘要
目的:研究新扶正除疫颗粒对感染流感病毒小鼠的抗病毒作用机制。方法:选择雌性BALB/c小鼠40只,并分成正常对照组(未攻毒)、病毒模型组、达菲对照组、新扶正除疫组,各10只,其中正常对照组、病毒模型组采用无菌生理盐水灌胃,达菲对照组采用达菲水溶液灌胃,新扶正除疫组采用新扶正除疫颗粒水溶液灌胃,持续给药7 d;同时病毒模型组、达菲对照组、新扶正除疫组于给药第3天开展FM1-6-E2滴鼻造模;四组于造模后的第3、6、9天制备脾脏细胞悬液,应用流式细胞仪对T细胞亚群进行检测、对比分析。结果:造模第3天病毒模型组小鼠的CD_4^+、CD_4^+/CD_8^+水平高于正常对照组,CD_8^+水平低于正常对照组,差异有统计学意义(P<0.05);达菲对照组、新扶正除疫组小鼠的CD_4^+、CD_4^+/CD_8^+水平低于病毒模型组,CD_8^+水平高于病毒模型组,差异有统计学意义(P<0.05);造模第6天病毒模型组小鼠的CD_3^+、CD_8^+水平高于正常对照组,CD_4^+、CD_4^+/CD_8^+水平低于正常对照组,差异有统计学意义(P<0.05);达菲对照组、新扶正除疫组小鼠的CD_3^+、CD_8^+水平低于病毒模型组,CD_4^+/CD_8^+水平高于病毒模型组,差异有统计学意义(P<0.05);造模第9d病毒模型组小鼠的CD_3^+、CD_8^+水平低于正常对照组,差异有统计学意义(P<0.05);而达菲对照组、新扶正除疫组小鼠的CD_3^+、CD_8^+水平高于病毒模型组,CD_4^+、CD_4^+/CD_8^+水平低于病毒模型组,差异有统计学意义(P<0.05)。结论:新扶正除疫颗粒能对感染流感病毒小鼠的细胞免疫功能进行调节,说明新扶正除疫颗粒调节T细胞亚群是其抗流感病毒主要作用机制之一。
Objective: To study the antiviral mechanism of New Fuzheng Chuyi Granule to influenza virus on mice infected with influenza virus. Methods: Forty female BALB/c mice were selected and divided into normal control group,virus model group,Duffy’s control group,and newly-remedy detoxification group. There were 10 of them,including normal control group and virus model group. Sterilized saline was given to the stomach. The Tamiflu control group was intragastrically administered with DAF aqueous solution. The New Fuzheng Chuyi Granule disease-free group was given intragastric administration of New Fuzheng Chuyi Granule. The administration was continued for 7 days. The virus model group,Duffy’s control group,and New Fuzheng Chuyi Granule were simultaneously administered. FM1-6-E2 intranasal injection was performed on the 3 rd day of the desegregation group,and spleen cell suspensions were prepared on the 3 rd,6 th and 9 th day after the injection in the four groups. Flow cytometry was used to detect T cell subsets. Results: The levels of CD4^+ and CD4^+/CD8^+ in the 3 rd-model virus model group was higher than that in the normal control group. The level of CD8^+ was lower than that in the normal control group,and the difference was statistically significant( P < 0. 05). The levels of CD4^+ and CD4^+/CD8^+ in the Duffy-treated control group and the newly-rooted detoxification group were lower than those in the virus model group. The level of CD8^+ was higher than that of the virus model group,and the difference was statistically significant( P < 0. 05). The levels of CD3^+ and CD8^+ in the 6 th-model virus model group were higher than those in the normal control group,and the levels of CD4^+ and CD4^+/CD8^+ were lower than those in the normal control group( P < 0. 05). The levels of CD3^+ and CD8^+ in the Tamiflu control group and the new Fuzheng group were lower than those in the virus model group. The level of CD4^+/CD8^+ was higher than that of the virus model group,and the difference was statistically significant(
