铜水杨酸菲咯啉络合物对三阴性乳腺癌裸鼠移植瘤生长的影响

摘要 目的探讨铜水杨酸菲咯啉络合物SODm对三阴性乳腺癌细胞株MDA-MB-231荷瘤裸鼠肿瘤的作用。方法对比细胞悬液种瘤法和组织插块种瘤法,选择最佳方式来构建三阴性乳腺癌细胞MDA-MB-231荷瘤裸鼠模型,采用HE染色检测荷瘤裸鼠模型是否构建成功。设置四个浓度梯度,即5μg/μl、1μg/μl、0.5μg/μl、0.1μg/μl,选择合适的SODm药物剂量浓度。实验组腹腔注射100μlSODm,对照组腹腔注射100μlDMSO,每间隔1d给药,共给药7次。给药后观察裸鼠的生长状态,并记录瘤块体积变化和裸鼠体重变化等。结果实验发现组织插块种瘤法成瘤快且成瘤率更高,故实验采用组织插块法构建MDA-MB-231裸鼠移植瘤模型。HE染色后镜下发现病理性核分裂象,证明裸鼠移植瘤模型构建成功。SODm给药浓度为0.5μg/μl,即每次给药量为50μg,该浓度剂量下的抑瘤效果最明显且安全性最高。实验组荷瘤裸鼠瘤块生长曲线明显低于对照组,且两组裸鼠体重无明显差别。结论插块法成功构建人类三阴性乳腺癌细胞系MDA-MB-231荷瘤裸鼠模型,体内实验证实了SODm对MDA-MB-231裸鼠移植瘤的生长具有抑制作用。 Objective To investigate whether the phenanthroline complex SODm of copper salicylic acid inhibit the tumor effect of three negative breast cancer cell line MDA-MB-231 bearing nude mice in vivo or not.Methods The breast cancer model of nude mice with MDA-MB-231 cancer cell line was built.SODm and DMSO were injected by intraperitoneal injection in the treatment group and control group,respectively.The doses were 100μl with 7 times,once every other day.The tumor volume and weight of those nude mice are observed and recorded after administration.Results It was found that the method of tumor tissue fragments inserting wass better with faster tumorigenicity and higher tumor rates.The MDA-MB-231 tumor-bearing nude mice model was constructed by this method.It was proved that the nude mouse model was constructed successfully by seeing irregular mitosis in HE staining.We choose the concentration of 0.5μg/μl of SODm,namely 50μg in every time,because of the better effect and high security.The tumor group growth curve in the treatment group was significantly lower than in the control group.But the weight fluctuation was no significantly different in the two groups.Conclusion The breast cancer cell line MDA-MB-231 tumor-bearing nude mice model is constructed by the method of tumor tissue fragments inserting.SODm can inhibit the tumor growth of breast cancer cell line MDA-MB-231 tumorbearing nude mice model in vivo.
