摘要
目的对3个品种药食两用欧李果原汁中的游离氨基酸含量进行差异比较分析。方法采用高效液相色谱-质谱联用技术(HPLC-MS/MS)测定欧李果原汁中的氨基酸。检测条件为:色谱柱:MSLab45AA-C18(150 mm×4.6 mm,5μm);流动相:A水为(含1%甲酸),B乙腈为(含1%甲酸);流速:1 mL·min^(-1);柱温:50℃。结果欧李果原汁中共检出18种游离氨基酸,其中京欧2号氨基酸总含量最高,达14 831.8μmol·L^(-1),显著高于农大4号(9414.4μmol·L^(-1))和内蒙古大欧李(4899.0μmol·L^(-1))。在京欧2号、农大4号和内蒙古大欧李3个品种的欧李果原汁中,均以脯氨酸含量最高,分别占氨基酸总含量的81.4%、82.4%、62.1%,品种间差异显著,以京欧2号为最高,表明该品种具有更强的抗逆性。结论欧李果原汁中含有丰富的必需氨基酸、支链氨基酸和药效氨基酸,且京欧2号显著高于其他两个品种。
Objective To compare and analyze the content of free amino acids in the juice of 3 medicinal fruits and edible fruits. Methods Using high performance liquid chromatography-mass spectrometry/mass spectrometry(HPLC-MS/MS) technology. We determined amino acids in Chinese dwarf cherry juice with OPA as the precolumn derivative reagent. The chromatographic column was MSLab45 AA-C18(150 mm×4.6 mm, 5 μm), the mobile phase included A water phase: water(1% formic acid), and B organic phase: acetonitrile(1-formic acid),the flow rate at 1 mL·min^-1 and column temperature at 50 ℃. Results Totally 18 free amino acids were detected in the juice of Chinese dwarf cherry. The total content of amino acids in the fruit juice of Jing Ou 2 was the highest, reaching 14 831.8 μmol·L^-1, much higher than that in Nong Da 4(9414.4 μmol·L^-1) and Inner Mongolia Chinese dwarf cherry(4899.0 μmol·L^-1). Proline content was the highest in the juice of all Jing Ou2, Nong Da 4 and Inner Mongolia Chinese dwarf cherry, and its contents of amino acids was 81.4%, 82.4% and62.1% respectively. Proline content of Jing Ou 2 was the highest among the three, indicating stronger resistance.Conclusion The juice of Chinese dwarf cherry has abundant essential amino acids, branched chain amino acids and effective amino acids. Jing Ou 2 has much higher percentage than the other two.
