miRNA-146a在桥本甲状腺炎合并乳头状甲状腺癌组织中的表达及其对甲状腺癌K1细胞的调控作用 被引量:11

摘要 目的探讨miRNA-146a在桥本甲状腺炎(HT)合并乳头状甲状腺癌(PTC)组织中的表达情况及其对甲状腺癌K1细胞增殖和侵袭能力的影响。方法选取215例甲状腺组织的标本,采用实时逆转录聚合酶链反应(RT-PCR)检测结节性甲状腺肿、HT、PTC、HT合并PTC组织中miRNA-146a的表达水平,并分析其与HT合并PTC患者临床特征的关系。将miRNA-146a inhibitor和阴性对照序列转染至甲状腺癌K1细胞中,分别作为miRNA-146a inhibitor组和NC组,CCK8法和克隆形成实验检测甲状腺癌K1细胞增殖和侵袭能力,蛋白质印迹法检测增殖细胞核抗原(PCNA)、细胞周期蛋白D1(cyclin D1)、基质金属蛋白酶9(MMP9)的表达水平。结果 HT、PTC、HT合并PTC组织中miRNA-146a的表达水平均高于结节甲状腺肿组织(P﹤0.05)。有淋巴结转移、肿瘤直径≥2 cm、TNM分期为Ⅲ~Ⅳ期的HT合并PTC患者miRNA-164a的表达水平均明显高于无淋巴结转移、肿瘤直径﹤2 cm、TNM分期为Ⅰ~Ⅱ期的患者(P﹤0.01)。miRNA-146a inhibitor组细胞miRNA-146a的增殖能力和克隆形成率均明显低于NC组细胞(P﹤0.01)。miRNA-146a inhibitor组细胞中PCNA、cyclin D1和MMP9蛋白的表达水平均明显低于NC组细胞(P﹤0.01)。结论 miRNA-146a在HT合并PTC组织中高表达,下调miRNA-146a的表达可通过抑制PCNA、cyclin D1和MMP9的表达抑制K1细胞的增殖和侵袭能力。 Objective To investigate the expression of miRNA-146a in Hashimoto’s thyroiditis(HT)combined with papillary thyroid carcinoma(PTC)and its effect on the proliferation and invasion ability of thyroid carcinoma K1 cells.Method The pathology specimens of 215 thyroid tissues were retrospectively analyzed,in which the relative expression of miRNA-146a was detected by fluorescent quantitative reverse transcription-polymerase chain reaction(RT-PCR),besides,the relationship between miRNA-146a expression and the clinicopathological parameters of patients with HT combined with PTC was analyzed.miRNA-146a inhibitor and negative control sequence were transfected into thyroid carcinoma K1 cells as miRNA-146a inhibitor group and NC group respectively,the proliferation and invasion ability of the cells were detected by CCK8 and colony formation assay,additionally,the protein levels of proliferating cell nuclear antigen(PCNA),cyclin D1(cyclin D1)and matrix metalloproteinase 9(MMP9)were detected by Western blot.Result Compared with nodular goiter tissues,the expression levels of miRNA-146a was significantly increased in HT,PTC and HT combined with PTC tissues(P<0.05).The expression of miRNA-146a in HT combined with PTC tissues was significantly higher in patients with lymph node metastasis,tumor diameter(≥2 cm)and TNM stage of Ⅲ-Ⅳ disease compared with those with no lymph node metastasis,tumor diameter(<2 cm)and TNM stage Ⅰ-Ⅱ disease(P<0.01).The proliferation and invasion ability of cells transfected with miRNA-146a inhibitor were decreased in comparison with NC group(P<0.01).The protein expression levels of PCNA,cyclin D1 and MMP9 proteins in miRNA-146a inhibitor group were significantly lower than that in NC group(P<0.01).Conclusion miRNA-146a is highly expressed in HT combined with PTC tissues.Down-regulation of miRNA-146a could reduce the proliferation and invasion ability of K1 cells of thyroid carcinoma by inhibiting expression of PCNA,cyclin D1 and MMP9.
