摘要
为比较蓝舌病病毒(BTV)cELISA和血清中和试验(SNT)检测方法的特点与关系,通过灭活、浓缩,制备抗原含量为1、5、10、50、100μg/mL的BTV-1型灭活疫苗。每个含量为1组,并设对照组,每组6只绵羊,分别在0、3周免疫。每周采血1次,共采血6次。对所采血样分别用cELISA和SNT方法进行抗体检测。cELISA检测结果显示,免疫后第1周,抗体阳性率达到80.00%,第4周全部免疫羊转为抗体阳性,抗体产生较为迅速;SNT检测结果显示,中和抗体阳性率从免疫后第1周的6.67%持续上升至第3周的36.67%,至第4周达到70.00%,中和抗体产生持续而缓慢。随着时间延长,2种检测方法的符合率由23.33%增加到76.67%。当绵羊同时产生中和抗体和cELISA抗体时,中和抗体效价与cELISA抑制率间有显著的线性相关关系(r=0.63),相关方程为y=0.38+0.05χ。分析认为,中和抗体出现的时间之所以晚于cELISA抗体,可能是因为与宿主细胞识别并吸附的主要部位VP7蛋白位于病毒衣壳的中间位置,其主要表面抗原位点暴露需要时间。
In order to compare the characteristics and explore the relationships between cELISA and the serum neutralization test(SNT)in detecting bluetongue virus(BTV),BTV-1 inactivated vaccines were prepared by inactivating and concentrating,and there were five kinds of concentrations:1,5,10,50 and 100 μg/mL, respectively. Each kind of concentration was considered as one group consisting of 6 sheep which were immunized in the 0th and 3rd week respectively. Also,the control group was set. Blood samples were collected once a week for 6 times,which were tested by cELISA and SNT respectively. The cELISA results showed that the positive rate of antibody reached 80.00% in the first week after vaccination,and turned to be 100% in the fourth week,showing that the antibodies were produced relatively rapid. While the results by SNT showed that the positive rate of neutralizing antibody increased from 6.67% in the first week to 36.67% in the third week after vaccination,and reached 70.00% in the fourth week,indicating that the neutralizing antibody appeared continuously and slowly. The coincidence rate of the two methods increased from 23.33% to 76.67% over time. When the neutralizing antibody and cELISA antibody appeared in sheep at the same time,there was an obvious linear correlation(r=0.63)occurred between the neutralizing antibody titer and the inhibition rate of cELISA,and the correlation equation was y=0.38+0.05χ. It was analyzed that,the reason why the neutralizing antibody appeared later than cELISA antibody was because VP7 protein recognized and adsorbed by host cells was located in the middle of viral capsid,thus it would take some time to expose its main surface antigen sites.
