摘要
为有效预防和控制蓝舌病病毒(BTV)感染,应用BTV-1型V863毒株,制备抗原含量分别为1、5、10、50、100μg/只份的BTV灭活疫苗进行绵羊免疫试验。设6个试验组(其中1组为空白对照),先后进行2次皮下免疫注射(2 m L/只),定期采集血清,利用微量血清中和试验方法(SNT)检测BTV-1型血清抗体水平,并应用感染BTV-1型的血毒进行攻毒试验,以评价疫苗免疫效果。结果显示:本研究制备的BTV-1型灭活疫苗能够刺激绵羊机体产生较好的中和抗体;2次免疫后,接种1、5、10、50、100μg/只份BTV灭活疫苗绵羊的抗体阳性率分别为66.7%、100%、83.3%、66.7%、50.0%,其中最高中和抗体滴度达362,攻毒后阴性率分别为66.7%、83.3%、83.3%、50.0%、66.7%。抗体滴度与保护率比较结果显示,当中和抗体滴度≥64时,病毒阴性率为100%。因此,10μg/只份的BTV灭活抗原含量可作为BTV灭活疫苗生产的推荐用量;BTV中和抗体滴度≥64可作为今后高效BTV灭活疫苗研发的依据。
In order to effectively prevent and control bluetongue virus(BTV),BTV-1 V863 strains were used to prepare five batches of inactivated vaccines containing viral antigens of 1,5,10,50 and 100 μg/head,and the immunological experiments were carried out.First,the experimented sheep were divided into 6 groups,including 1 blank control group.Then the subcutaneous immunizations were performed twice towards the sheep(2 m L per sheep),the serum samples were collected regularly and antibody detection was conducted by sero-neutralization test(SNT)method.Eventually,the blood carrying BTV-1 viruses was used to carry out challenge infection to evaluate the immune effect of vaccines.Results showed that the inactivated vaccines of BTV-1 prepared in this study could stimulate the sheep to produce neutralizing antibodies.After twice vaccination,the positive rates of antibodies in different sheep groups injected with vaccines of 1,5,10,50 and 100 μg/head pathogens were 66.7%,100%,83.3%,66.7%,50.0%,respectively.And the highest level of neutralizing antibody titer could reach to 362.After the challenge infection,the virus negative rates of the five sheep groups were 66.7%,83.3%,83.3%,50.0%,66.7%,respectively.The comparison of antibody titer and protection rate showed that when the antibody titer was above 64,the negative rate of virus was 100%.Therefore,vaccines containing 10 μg/head of viral antigens could be used as the recommendation dosage during production,and the antibody titer of over 64 could be regarded as the basis for effective research and development of BTV inactivated vaccines in the future.
