摘要
肥胖已成为世界的流行病并可引发与肥胖相关的疾病,而肥胖的产生与脂肪细胞的增殖及分化密切相关。本研究采用MTT法测定了石榴皮提取物及鞣花酸对融合期人脂肪干细胞增殖的抑制活性,采用油红染色法观察分化期脂肪干细胞的形态学变化及测定甘油三酯的相对含量。研究表明,随着石榴皮多酚提取物和鞣花酸浓度的增加,脂肪干细胞的增殖和分化出现呈剂量效应的抑制作用。当石榴皮提取物质量浓度为25μg·m L-1时,对脂肪干细胞增殖和分化的抑制率分别为31. 87%、45. 88%。鞣花酸质量浓度为4. 5μg·m L-1时,对脂肪干细胞增殖和分化的抑制率分别为47. 23%、44. 96%。说明石榴皮多酚提取物及鞣花酸可有效抑制人脂肪干细胞增殖和分化为成熟脂肪,减少甘油三酯的累积。鞣花酸与石榴皮多酚提取物相比具有更强的抑制作用,提示其具有潜在的预防肥胖作用。
Obesity has become an epidemic in the world and can cause diseases related to obesity,and the production of obesity is closely related to the proliferation and differentiation of adipocytes.In this study,MTT method was used to determine the inhibitory activity of pomegranate peel extract(PoPE)and Ellagic acid(EA)on the proliferation of human adipose-derived stem cells(h ADSCs)during fusion.The morphological changes and the relative content of triglycerides of differentiated h ADSCs were observed by Oil Red-O staining.Studies have shown that with the increase of the concentration of PoPE and EA,the proliferation and differentiation of h ADSCs have a dose-effect inhibitory effect.When the concentration of PoPE was 25μg·mL^-1,the inhibition rates of proliferation and differentiation on h ADSCs were 31.87%and 45.8%,respectively.When the concentration of EA was 4.5μg·mL^-1,the inhibition rates on proliferation and differentiation of h ADSCs were 47.23%and 4.96%,respectively.It indicated that the PoPE and EA can effectively inhibit the proliferation and differentiation of h ADSCs into mature adipocytes and reduce the accumulation of triglycerides.The EA has a stronger inhibitory effect than the PoPE,suggesting that EA in pomegranate plays a potential role on obesity prevention.
