牛源致病性大肠埃希菌K99对小鼠肠绒毛生长和肠三叶因子mRNA表达的影响 被引量:3

Effects of Pathogenic E. coli K99 from Bovine on Growth of Intestinal Villi and Expression of ITF mRNA in Mice

摘要 通过研究大肠埃希菌对小鼠肠绒毛生长和肠三叶因子mRNA表达的影响,探讨牛源致病性大肠埃希菌感染与肠黏膜上皮细胞再生的相互关系。选取Balb/c小鼠,分成空白对照组、Brdu组(Brdu)和E.coli感染组(Brdu+E.coli),并于不同时间点(6、12、24、30、36、48、60h)采集小鼠十二指肠、空肠和回肠组织。用HE染色观察病理组织学变化,并测量肠绒毛长度和隐窝深度;用免疫组化染色检测肠黏膜上皮细胞中Brdu阳性信号;用PAS染色检测杯状细胞数;用real-time PCR检测肠黏膜组织中ITF mRNA表达水平。结果显示,空白对照组和Brdu组肠组织结构无明显变化。E.coli感染小鼠肠组织6h^36h后,十二指肠、空肠绒毛萎缩或崩解成碎片脱落,隐窝变深;其他时间未见明显变化。E.coli感染小鼠后6h^36h,Brdu阳性信号在上皮细胞中的移动距离极显著高于Brdu组(P<0.01)。E.coli感染小鼠后6h^60h,十二指肠、空肠、回肠中的杯状细胞数均显著低于空白对照组和Brdu组(P<0.01)。E.coli感染小鼠后6h^36h,小肠组织中ITF mRNA表达极显著低于空白对照组和Brdu组(P<0.01)。牛源致病性E.coli感染小鼠后,既可引起小肠黏膜上皮细胞的损伤和抑制ITF mRNA表达,也能够促进小肠绒毛上皮细胞的生长。 To study the relationship between pathogenic E.coli and regeneration of intestinal epithelial cells through investigating the effects of E.coli on growth of intestinal villi and mRNA expression of intestinal trefoil factor (ITF) in mice,intestinal tissues including duodenum,jejunum,ileum in mice,which were divided into control group,Brdu group (Brdu) and E.coli -infection group ( E.coli +Brdu),were collected at 6,12,24,30,36,48,60 h post treatment.Histopathological lesions,the lengths of intestinal villi and depth of crypts,the location of Brdu positive cells and the population of goblet cells were measured by histopathological observation,immunohistochemical staining,PAS staining and qRT-PCR,respectively.The results showed that atrophied villi and cell fragments were observed in intestine tissues after treatment for 6 h-36 h in E.coli group,while there was no changes in control and Brdu groups.Immunohistochemistry results revealed significantly higher migration length in E.coli group than in control and Brdu groups during 6 h-36 h after E.coli infection ( P <0.01).The population of goblet cells decreased dramatically after E.coli infection from 6 h to 60 h.The mRNA expression of ITF in E.coli group was significantly lower than that in control and Brdu groups from 6 h to 36 h ( P <0.01).In this study,it is demonstrated that intestinal mucosal epithelial cells injury and the growth of intestinal villus epithelial cells were induced by pathogenic E.coli ,which inhibited the expression of ITF mRNA.
