摘要
目的:本文旨在研究五味子乙素对肝癌细胞HCCLM3凋亡、侵袭及血管新生的影响。方法:MTT检测细胞存活力。流式细胞术分析细胞凋亡。Transwell检测细胞侵袭。蛋白印迹检测Bcl 2相关蛋白X(Bax)、B细胞淋巴瘤(Bcl-2)、基质金属蛋白酶2(MMP-2)、MMP-9、血管内皮细胞生长因子X(VEGF-A)、表皮生长因子(EGF)和成纤维细胞生长因子(b FGF)表达。结果:低浓度(<20μmol/L)五味子乙素对HCCLM3细胞存活力无明显影响。高浓度(>20μmol/L)五味子乙素会降低细胞存活力。与DMSO组相比,五味子乙素(10和20μmol/L)组细胞凋亡率明显升高,Bax表达增强,Bcl-2表达降低(P<0. 05)。而且,五味子乙素(5、10、20μmol/L)组细胞侵袭及MMP-2和MMP-9表达明显低于DMSO组(P<0. 05)。与DMSO组相比,五味子乙素(5、10、20μmol/L)组VEGF-A,EGF和b FGF表达明显下降(P<0. 05)。结论:五味子乙素可诱导肝癌细胞HCCLM3凋亡,降低细胞侵袭及血管新生。
Objective:To research the effect of schisandrin B on the apoptosis,invasion and angiogenesis of liver cancer cells HCCLM3.Methods:Cell viability was detected by MTT.Apoptosis was measured by flow cytometry.Cell invasion was tested through Transwell.The expression of B cell lymphoma 2(Bcl-2),matrix metalloprotein 2(MMP-2),MMP-9,vascular endothelial growth factor A(VEGF-A),epidermal growth factor(EGF)and basic fibroblast growth factor(bFGF)was detected by Western blot.Results:Low concentration(<20μmol/L)schisandrin B had no effect on cell viability of HCCLM3 cells.The cell viability was reduced by high concentration( >20μmol/L)schisandrin B.Compared with DMSO group,the apoptosis rate and expression of Bax in schisandrin B(10,20μmol/L)groups were enhanced with decreased expression of Bcl-2(P<0.05).Moreover the cell invasion and expression of MMP-2 and MMP-9 in schisandrin B(5,10,20μmol/L)groups were lower than DMSO group(P<0.05).Compared with DMSO group,the expression of VEGF-A,EGF and bFGF in schisandrin B(5,10,20μmol/L)groups was reduced(P<0.05).Conclusion:Schisandrin B induces apoptosis and decreases cell invasion and angiogenesis of HCCLM3 cells.
