摘要
目的:检测长链非编码RNA(long non-coding RNAs,lncRNAs)肺癌相关转录本1(lung cancer associ-ated transcript 1,lucat1)在胃癌组织和细胞中的表达,探讨lucat1与胃癌临床病理特征及预后的关系。方法:利用RT-PCR检测胃癌组织、细胞及配对癌旁组织、上皮细胞中lncRNA lucat1的表达;利用单因素方差分析判断lucat1与胃癌患者临床病理特征之间的关系;利用生存分析曲线描述lucat1与胃癌患者术后5年生存率的关系;利用Cox风险比例模型分析lucat1与胃癌患者预后的关系。结果:lncRNA lucat1在胃癌组织、细胞中表达水平高于癌旁组织和胃上皮细胞; lucat1的表达与胃癌TNM分期相关(P <0. 05); lucat1高表达的胃癌患者术后5年生存率低于低表达的患者,lucat1表达是胃癌患者独立的预后因素。结论:lncRNA lucat1在胃癌组织和细胞中表达升高,并与胃癌患者生存及预后密切相关。
Objective: To investigate the expression level of long non-coding RNA (lncRNA) lung cancer associated transcript 1 (lucat1) in gastric cancer,and evaluate its prognostic value in gastric cancer. Methods: Gastric cancer cells and gastric epithelial cell were tested in this study.Cancer tissue as well as adjacent normal tissue from patients diagnosed with gastric cancer were also harvested in this study.Expression of lucat1 was measured by RT-PCR.ANOVA analysis was performed to evaluate the association between lucat1 and clinical manifestation of gastric cancer patients.K-M survival analysis was utilized to determine correlation of lucat1 expression with 5-year survival.Cox's proportional hazards regression models were applied to analyze the relationships between lucat1 and prognosis of gastric cancer patients. Results: Lucat1 was upregulated in gastric cancer tissues and cells compared with normal tissues and cell.Expression of lucat1 was closely associated with TNM stage of gastric cancer ( P <0.05).Expression of lucat1 was negatively correlated with 5 -years survival rate.Lucat1 is an independent prognostic risk factor of gastric cancer. Conclusion: lncRNA lucat1 is upregulated in gastric cancer and is negatively associated with outcome of gastric cancer.
