摘要
目的观察重症肌无力(MG)患者血清microRNA-146a水平变化,并采用生物信息学方法分析microRNA-146a在MG发病中的分子调控网络。方法选取初诊MG患者108例(MG组)及同期体检健康者50例(对照组),采用RT-PCR法检测血清microRNA-146a。采用UCSC基因组在线工具分析microRNA-146a在人类基因组中的位置及保守性,采用Targetscan和Co MeTa数据库预测microRNA-146a的交集靶基因,采用DAVID数据库分析microRNA-146a靶基因的功能富集(GO富集)和信号通路富集(KEGG Pathway)。结果 MG组血清microRNA-146a相对水平为2. 13±0. 63,高于对照组的0. 86±0. 38(P <0. 05); microRNA-146a核苷酸序列具有高度保守性,其潜在靶基因有ERBB4、NUMB、CD80、TRAF6、IRAK1等88种; GO富集结果显示,microRNA-146a靶基因功能主要富集于细胞增殖调控、神经元分化、磷代谢反应、免疫炎症反应、细胞因子合成等过程; KEGG Pathway结果显示,microRNA-146a靶基因主要富集于Toll样受体信号通路、神经递质调节信号通路、EB信号通路等。结论 MG患者血清microRNA-146a水平升高,其可通过作用于多条信号通路参与MG的发病。
Objective To observe the changes in levels of microRNA-146a in the serum of patients with myasthenia gravis(MG),and to analyze the molecular regulatory network of microRNA-146a in the pathogenesis of MG by bioinformatics.Methods One hundred and eight patients who was first diagnosed MG(MG group),and 50 healthy persons in the same period(control group)were selected,and then the serum microRNA-146a was tested by RT-PCR.The UCSC genome online tools were used to analyze the position and conservatism of microRNA-146a in the human genome.Targetscan and CoMeTa database was used to predict the inter-target gene of microRNA-146a.DAVID database was used to analyse the functional enrichment(GO enrichment)and signal pathway enrichment(KEGG Pathway)of target gene in microRNA-146a.Results The serum microRNA-146a in the MG group was 2.13±0.63,which was higher than that(0.86±0.38)of control group(P<0.05).Nucleotide sequence was highly conserved in microRNA-146a,with 88 types of potential target genes ERBB4,NUMB,CD80,TRAF6,IRAK1,etc.GO analysis showed that the function of microRNA-146a target gene mainly focused on the synthesis process of cell proliferation regulation,neuronal differentiation,phosphorus metabolism reaction,immune inflammation,and cytokines,etc.KEGG Pathway showed that microRNA-146a target gene mainly focused on Toll receptor signaling pathway,neurotransmitter regulatory signaling pathway,EB signaling pathway,and other signaling pathways.Conclusion The level of microRNA-146a is elevated in the serum of MG patients,which can participate in the pathogenesis of MG by acting on multiple signaling pathways.
