miR-210、BNIP3在大鼠脑缺血再灌注损伤的表达及意义 被引量:9

Expressions and Significances of miR-210 and BNIP3 Proteins in Rats with Cerebral Ischemia-reperfusion Injury

摘要 目的探讨miR-210、BNIP3蛋白在大鼠脑缺血再灌注损伤中表达情况。方法 40只成年雄性SD大鼠随机分为假手术组、大脑中动脉脑缺血再灌注(MCAo)后6、12、24 h组,每组10只。计算4组大鼠脑梗死体积,测定4组脑皮质区miR-200 mRNA、BNIP3 mRNA及BNIP3蛋白表达情况。结果 MCAo后6、12、24 h组大鼠脑梗死体积大于假手术组,且随缺血时间延长而增大(P <0. 05)。MCAo后6、12、24 h组缺血脑皮质区miR-210、BNIP3 mRNA及BNIP3蛋白相对表达水平高于假手术组,且MCAo组随缺血时间延长而升高(P <0. 05)。结论脑缺血再灌注损伤后miR-210表达水平上调,与BNIP3 mRNA表达趋势一致,miR-210与BNIP3可能协同参与脑缺血再灌注损伤过程。 Objective To investigate expressions of miR-210 and Bcl-2/adenovirus EIB-interacting protein 3(BNIP3)proteins in rats with cerebral ischemia-reperfusion injury.Methods A total of 40 adult male SD rats were randomly divided into sham operation group,6h,12h and 24h after middle cerebral artery occlusion(MCAo)groups(n=10 for each group).Areas of cerebral infarction were calculated,and expressions of miR-200 mRNA,BNIP3 mRNA and BNIP3 protein in rats'cerebral cortex were detected among 4 groups.Results Areas of cerebral infarction in 6h,12h and 24h after MCAo groups were larger than that in sham operation group,and the areas were increased with prolonging ischemic time(P<0.05).Compared with those in sham operation group,relative expressions of miR-210 mRNA,BNIP3 mRNA and BNIP3 protein in cerebral cortex were significantly higher in 6h,12h and 24h after MCAo groups(P<0.05),and the levels in MCAo groups were increased with prolonging ischemic time(P<0.05).Conclusion The miR-210 expression is increased after cerebral ischemia-reperfusion injury,which is consistent with expression of BNIP3 mRNA,and therefore miR-210 and BNIP3 may be involved in process of cerebral ischemia-reperfusion injury in rats.
