湖北省烟草赤星病长柄链格孢遗传多样性分析 被引量:5

摘要 为从分子水平揭示烟草长柄链格孢菌群体遗传学特性,本研究采用ISSR分子标记方法对十堰、襄阳、恩施、宜昌4个烟区的34株长柄链格孢菌进行了遗传多样性、遗传结构与分化以及种群间遗传变异分析;其中Nei’s多样性指数为0.1118~0.2061,Shnanon信息指数为0.1972~0.3177,表明长柄链格孢在不同地理种群间具有丰富的遗传多样性;遗传距离分析表明,十堰和襄阳,恩施与宜昌的菌株亲缘关系较近;居群每代迁移数(Nm)为3.2740> 1,表明不同地理的种群间存在基因流动;利用NTSYS软件按UPGMA方法聚类分析,当相似性系数设定为0.79时,可将34个供试菌株分为4个类群,但菌株并没有按地理来源聚为不同的组,表明菌株之间的遗传多样性与其地理来源无明显的相关性。 To reveal genetic characteristics of Alternaria longipes at molecular level,genetic diversity,genetic structure,differentiation and genetic variation of 34 strains of Alternaria longipes from four tobacco growing areas of Shiyan,Fuyang,Enshi and Yichang were analyzed by ISSR molecular marker method.Nei’s diversity index and Shnanon’s information index were ranged from 0.1118 to 0.2061 and 0.1972 to 0.3177 respectively,which indicated that there was abundant genetic diversity among A.longipes from different geographic areas.Genetic distance analysis showed that strains between Shiyan and Xiangyang and those between Enshi and Yichang were closely related.Population migration per generation(Nm)was 3.2740>1,indicating that there were gene flows among fungus from different geographic areas.According to UPGMA cluster analysis using NTSYS software,the 34 isolates can be classified into 4 groups when the similarity coefficient was set to 0.79.However,these strains can not be grouped according to geographical origin,indicating that there was no significant correlation between genetic diversity and geographical origin.
