摘要
以彩色马蹄莲‘卡格丽’球茎为外植体,探讨其组织培养技术。结果表明,经75%酒精处理1 min,再用0.1%HgCl_2消毒20 min,可建立彩色马蹄莲‘卡格丽’无菌体系。培养基优化试验表明,‘卡格丽’最适继代培养基为:MS+1.5 mg·L^(-1)6-BA+0.2 mg·L^(-1)NAA+30 g·L^(-1)蔗糖;最适生根培养基为:1/2MS+0.6 mg·L^(-1)NAA+0.2 g·L^(-1)活性炭+15 g·L^(-1)蔗糖。
The bulbs of Zantedeschia hybrida‘Calgary’were used as the explants to explore the tissue culture technique.The results showed that the sterile system of‘Calgary’could be established by treating the explants with75%ethanol for1min and then disinfecting with0.1%HgCl2for20min.The medium optimization experiment showed that the optimum subculture medium for‘Calgary’was:MS+1.5mg·L^-16-BA+0.2mg·L^-1NAA+30g·L^-1sucrose.The optimum rooting medium was:1/2MS+0.6mg·L^-1NAA+0.2g·L^-1activated carbon+15g·L^-1sucrose.
