摘要
目的探讨羟基红花黄色素A(HSYA)对柯萨奇病毒性(CVB3)心肌炎小鼠TNF配体相关分子-1A(TL1A)及干扰素-γ(IFN-γ)表达的影响。方法 Wistar小鼠90只随机分为正常组、模型组及治疗组,腹腔注射CVB3诱发心肌炎小鼠模型。测定心肌肥厚指数,HE染色观察心肌变化,检测血清TL1A和IFN-γ水平。结果模型组小鼠血清TL1A及IFN-γ水平均较正常组显著升高(P <0. 05);与模型组相比,治疗组血清TL1A及IFN-γ水平显著降低(P <0. 01),但仍高于正常组(P <0. 05)。模型组心肌肥厚指数和心脏质量显著高于正常组(P <0. 01),经过治疗后,治疗组的心肌肥厚指数和心脏质量明显降低,显著低于模型组(P <0. 05),但仍略高于正常组(P <0. 05)。各组小鼠心肌组织病理改变,正常组小鼠心肌细胞形态正常,未见水肿等病变;模型组小鼠心肌细胞排列紊乱,心肌横纹不清,大量炎性细胞浸润;治疗组心肌纤维形态和排列基本正常。结论 HSYA可能通过调节血清TL1A、IFN-γ水平保护CVB3心肌炎的心肌细胞,减轻CVB3所诱导的心肌损伤。
Objective To explore the Effects of hydroxysafflor yellow A(HSYA)on serum TL1A and IFN-γex-pressions in the mice with CVB3myocarditis.Methods a total of90Wistar mice were randomized into a normal group,a model group and a treatment group.The intraperitoneal injection of CVB3was used to induce the myocarditis models in the mice.The myocardial hypertrophy index was determined.Using HE staining,the myocardial change was observed.The TL1A and IFN-γexpressions were determined in the serum.Results The TL1A and IFN-γexpressions in the model group were increased significantly as compared with the normal group(P<0.05).Compared with the model group,the ex-pressions in the treatment group were reduced significantly(P<0.01),but they were still higher than the normal group(P<0.05).The myocardial hypertrophy index and heart mass in the treatment group were reduced obviously,which were lower significantly than the model group(P<0.05),but still higher than the normal group(P<0.05).In each group,the myocardial tissue changed pathologically.The myocardial cells were normal in morphology in the normal group and no ede-ma was found.In the model group,the myocardial cells were disarranged,the myocardial stripes unclear and the inflamma-tory cells infiltrated in a large amount.In the treatment group,the myocardial fibers were normal basically in morphology and arrangement.Conclusion HSYA protects myocardial cells and alleviates the induced myocardial injury probably through regulating serum TL1A and IFN-γexpressions in the mice with CVB3 myocarditis.
