摘要
目的观察苦豆碱对异丙肾上腺素所致大鼠心肌肥厚的保护作用。方法雄性Sprague-Dawley大鼠,随机分为5组:正常对照组、模型组及10、20、40mg·kg^(-1)苦豆碱组,连续灌胃给药14 d。于第8天皮下注射异丙肾上腺素(ISO,5mg·kg^(-1),连续7 d)建立大鼠心肌肥厚模型。观察苦豆碱对心肌肥厚大鼠左心室内压最大上升速率(+LVdp/dtmax)、左室收缩压(LVSP)、左心室内压最大下降速率(-LVdp/dtmax)、平均动脉压、心率、心肌肥厚指数、心肌组织形态结构的影响;分离血清,检测苦豆碱对心肌肥厚大鼠心肌肌钙蛋白(cTnⅠ)及丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)等的影响。结果与模型组比较,20和40mg·kg^(-1)苦豆碱可降低大鼠全心湿质量、左心室湿质量、全心湿质量/体质量比与左心室湿质量/体质量比,升高+LVdp/dtmax、-LVdp/dtmax,降低LVEDP(P<0.05);苦豆碱可减轻大鼠心肌细胞肥大、纤维化、间质水肿和炎细胞浸润等ISO导致的心肌组织病理学结构的异常改变;同时降低c TnⅠ水平(P<0.01)。此外,20mg·kg^(-1)和40mg·kg^(-1)苦豆碱能提高血清中SOD和GSH-PX活性,并降低MDA的含量(P<0.05)。结论苦豆碱对ISO所致大鼠心肌肥厚具有保护作用,其作用机制可能与提高大鼠的抗氧化能力有关。
Objective To investigate the protective effect of aloperine on isoproterenol-induced cardiac hypertrophy in rats and its mechanisms.Methods Male Sprague Dawley rats were given aloperine(10,20,40 mg·kg^(-1)) orally for 14 days.Myocardial hypertrophy model was induced in rats by subcutaneous injection of isoproterenol(5 mg·kg^(-1))at the 8 th day for 1 week.The haemodynamic parameters,the body weight,myocardial hypertrophy indexes and histomorphology were checked to evaluate the effect of aolperine on isoproterenol-induced myocardial hypertrophy in rats.In addition,levels of cardiac troponinsⅠ,superoxide dismutase,malondialdehyde and glutathione peroxidase in serum were measured.Results Compared with model group,oral administration of aloperine considerably decreased LVEDP and increased LVdp/dtmax on myocardial hypertrophy rats.Aloperine treatment significantly decreased the myocaridial hypertrophy indexes and attenuated myocardial necrosis,interstitial edema and inflammatory cell infiltration.Moreover,aloperine decreased the level of cardiac troponins Ⅰ and increased antioxidant enzyme activity to different degrees and enhanced the activity of superoxide.Conclusion These results demonstrate that aloperine exerts protective effects on isoproterenol-induced myocardial hypertrophy in rats,which is likely to be mediated through increasing antioxidant enzyme activities and normalization the balance of oxidation and antioxidation.
