摘要
目的通过不可逃避电足刺激建立创伤后应激性障碍(PTSD)动物模型,观察葛根素对PTSD模型小鼠行为学的影响。方法 60只雄性ICR小鼠随机分为正常对照组,模型组,舍曲林组,葛根素低、中、高剂量组。除正常对照组外其余各组均给予PTSD造模,造模后正常对照组、模型组每天灌胃生理盐水,舍曲林组和葛根素低、中、高剂量组分别给予15 mg/kg舍曲林及7. 5 mg/kg、15 mg/kg和30 mg/kg葛根素灌胃。试验的第5、10、17天检测僵住行为。结果第5天检测结果显示舍曲林组不能产生抗PTSD样效应,但葛根素中、高剂量组均可逆转小鼠PTSD样僵住时间百分比显著上升。第10、17天结果显示舍曲林组和葛根素中、高剂量组均可逆转小鼠僵住时间百分比显著上升。结论葛根素对不可逃避电足建立PTSD模型僵住行为有良好的改善作用。
Objective To observe the ethological effects of puerarin on the model mice with posttraumatic stress disorder(PTSD)induced by inescapable electric foot stimulation.Methods A total of60male ICR mice were randomized into a normal control group,a model group,a sertraline group,the low-dose,middle-dose and high-dose groups of puerarin.Except normal control group,PTSD models were prepared in the rest groups.After modeling,the physical saline was used for gavage in the normal control group and the model group every day.In the sertraline group,the low-dose,middle-dose and high-dose groups of puerarin,sertraline15mg/kg,puerarin,7.5mg/kg,15mg/kg and30mg/kg were used in gavage separately.On day5,10and17of the experiment,the freezing behavior was evaluated.Results On day5,in the sertraline group,the effect for anti-PTSD was not presented,but in the puerarin middle-dose and high-dose groups,the significant increase of PTSD-like freezing time percentage was reversed in the mice.On the day10and17,in the sertraline group,the middle-dose and high-dose groups of puerarin,the significant increase of freezing time percentage was reversed in the mice.Conclusion Puerarin has the good improvement effects on(PTSD induced by inescapable electric foot stimulation.The effect mechanism needs to be explored.
