绵羊ADPN基因和PTGS2基因多态性与产羔数相关性研究

摘要 实验旨在研究ADPN基因和PTGS2基因与湖羊和杜泊羊产羔数的相关性,采用直接测序法检测65只湖羊和60只杜泊羊的ADPN基因的第3外显子以及PTGS2基因的第3外显子到第5外显子的多态性,并分析其与产羔数的相关性。结果表明:湖羊在ADPN基因第3外显子的133 bp处突变的密码子由GAT转变成AAT,并使编码的氨基酸由D(天冬氨酸)转变成N(天冬酰胺);在594bp处突变的密码子由AGA转变为AAA,使编码的氨基酸由R(精氨酸)转变为K(赖氨酸);湖羊在PTGS2基因第3外显子的77 bp位点的密码子由AAA转变为CAA,并使编码的氨基酸由K(赖氨酸)转变为Q(谷氨酰胺);但在杜泊羊群体中并没有发现基因的突变。因此推测ADPN基因和PTGS2基因可能是引起湖羊高繁殖力的基因。 The experiment aimed to study the relationship between ADPN and PTGS2 genes and litter size of Hu sheep and Dorper sheep.A direct sequencing method was used to detect the polymorphism of the 3rd exon of the ADPN gene and the 3rd exon to 5th exon of the PTGS2 gene,the genes tested were from 65 Hu sheep and 60 Dorper sheep.The correlation with litter size was analyzed.The results showed that the mutant codon was appeared at 133 bp of exon 3 of the ADPN gene,the mutant codon changed from GAT to AAT and the encoded amino acid was changed from D(Aspartic acid)to N(asparagine);the mutant codon at 594 bp changed from AGA to AAA and the encoded amino acid was changed from R(arginine)to K(lysine);the mutant codon appeared at 77 bp of exon 3 changed from AAA to CAA and the encoded amino acid was changed from K(lysine)to Q(glutamine).However,no mutations were found in the population of Dorper sheep.Therefore,it is speculated that the ADPN gene and the PTGS2 gene may be the genes that cause the high fertility of the Hu sheep.
