MRAS基因多态性与急性心肌梗死及Syntax评分的相关性分析

摘要 目的分析肌RAS原癌基因同源基因(MRAS) rs3755751位点多态性与急性心肌梗死(AMI)及Syntax评分的关系。方法选择2006年6月—2015年12月常州市武进人民医院心内科AMI患者248例作为研究对象,采用病例—对照研究,应用基因测序方法检测AMI患者248例(AMI组)和非冠心病者310例(对照组)的MRAS基因rs3755751位点多态性。采用Syntax评分软件进行Syntax评分。结果 AMI组CC、CT和TT基因型频率与对照组比较,差异无统计学意义(χ~2=4. 231,P=0. 121)。进一步构建效应模型,CC vs.(CT+TT)和TT vs.(CT+CC),AMI组与对照组基因型差异无统计学意义(P=0. 056和0. 934)。进一步多元Logistic回归显示,在调整其他危险因素后,CC基因型与AMI无显著性关联(P=0. 796)。CC基因型组Syntax评分明显低于CT及TT基因型组(CC vs.CT:P=0. 006; CC vs. TT:P=0. 007),而CT与TT 2组间Syntax评分无显著性差异(P=0. 167)。结论 MRAS基因rs3755751位点多态性与AMI发病无显著性关联,但AMI患者CC基因型的Syntax评分明显低于CT和TT基因型。 Objective To investigate the association of rs3755751 loci polymorphism in the muscle RAS oncogene homolog gene(MRAS)with acute myocardial infarction(AMI)and Syntax score.Methods A case-control study was conducted.The MRAS gene rs3755751 loci polymorphism was analyzed in 248 patients with AMI(AMI group)and 310 control subjects free from coronary heart disease(control group)by gene sequencing method.Syntax scoring software was used to score Syntax.Results The frequencies of CC,CT and TT genotypes in AMI group were not significantly different from those in the control group(χ2=4.231,P=0.121).Further construction of an effect model,CC vs.(CT+TT)and TT vs.(CT+CC),there was no significant difference between the AMI group and control group(P=0.056,P=0.934).Furthermore,multivariate Logistic regression showed that there was no significant association between CC genotype and AMI after adjustment for other risk factors(P=0.796).The Syntax score of the CC genotype group was significantly lower than those of the CT and TT genotype groups(CC vs.CT:P=0.006,CC vs.TT:P=0.007),but there was no significant difference between the CT group and TT group(P=0.167).Conclusion There is no significant relationship between The MRAS gene rs3755751 loci polymorphism and the incidence of AMI,but the Syntax score of CC genotype is significantly lower than those of CT and TT genotypes.The findings further supplement experimental data for MRAS gene study of the coronary disease.
