稳恒直流电场对鸡胚绒毛尿囊膜血管生长的影响 被引量:1

摘要 背景:组织工程骨因其微创、安全、精确的特性,已成为恢复颌面部骨缺损及牙周组织的重要手段。目的:以鸡胚绒毛尿囊膜血管为模型,加载不同强度电场,检验血管生长相关参数,研究电场对组织工程骨预血管化的作用,以期得到促进组织工程骨预血管化的方法,增加缺损修复成功率。方法:使用孵化至第8天的鸡胚建立鸡胚绒毛尿囊膜血管模型(Inshell模型),将模型随机分为A、B、C、D、E 5个组,分别加载0,-0.75,-1.5,-3,+1.5 V的不同大小及不同方向的稳恒直流电场,在加电0,12,24,36,48 h拍摄相关图片,使用Image J测量相关血管数据,观察指标为血管平均长度及血管生长角度,使用SPSS 22.0分析结果。结果与结论:(1)加载不同大小电场以后,A组平均血管长度增长速率为0.012 mm/h,B组为0.023 mm/h,C组为0.047 5 mm/h;(2)使用独立样本t检验进行比较,加载-1.5 V电压48 h后的绒毛尿囊膜血管平均长度为(4.22±0.436)mm/mm^2,加载0 V电压48 h后为(3.21±0.31)mm/mm^2,两者差异有显著性意义(P <0.01);(3)48 h后,加载-1.5 V电压血管角度为(21.36±20.16)°,加载0 V电压则为(82.17±50.44)°,差异有显著性意义(P <0.05);(4)D、E2组电极周围的鸡胚绒毛尿囊膜发生变性,鸡胚死亡;(5)提示适宜大小范围内的负电促进血管生长,同时改变血管生长角度,证明稳恒直流电场对鸡胚绒毛尿囊膜血管有明显影响。 BACKGROUND:Tissue-engineered bone has become an important means of repairing maxillofacial bone defects due to its minimally invasive,safe and accurate characteristics.OBJECTIVE:The model of chick chorioallantoic membrane was loaded with different intensities of electric field,to detect the blood vessel growth-related parameters,and to explore the effect of electric field on force-vascularization of tissue-engineered bone,in order to search for a method that can promote force-vascularization of tissue-engineered bone,thus increasing the repair success rate.METHODS:The model of chick chorioallantoic membrane(Inshell model)was established using the chicken embryos hatched for 8 days.The models were randomized into A,B,C,D,and E groups,followed by loaded with 0,-0.75,-1.5,-3 and+1.5 V and steady direct current electric fields at different directions.The images were taken at 0,12,24,36 and 48 hours.The average blood vessel length and growth angle were measured by Image J,and analyzed on SPSS 22.0 software.RESULTS AND CONCLUSION:After loaded with different currents of electric fields,the average growth rate of vessel length in the groups A,B and C was 0.012,0.023,and 0.047 5 mm/h,respectively.Independent sample t test results revealed that after loaded with-1.5 and 0 V for 48 hours,the average length of blood vessel was(4.22±0.436)and(3.21±0.31)mm/mm2,respectively,and the difference was significant(P<0.01).After loaded with-1.5 and 0 V for 48 hours,the angle of blood vessels was(21.36±20.16)°and(82.17±50.44)°,respectively,and the difference was significant(P<0.05).The chick chorioallantoic membrane around the electrode in the groups D and E degenerated and chick embryos died.These findings indicate that appropriate negative electricity promotes the growth of blood vessels,and changes the angle of blood vessel growth,suggesting that steady direct current electric field greatly affects chick chorioallantoic membrane.
