摘要
目的:研究急性胰腺炎模型中胰腺组织中F-actin、RegⅢ的水平变化情况。方法:把SD大鼠分成3组,每组15只,分别是模型组、假手术组及空白组,急性胰腺炎的模型组是利用3%牛磺胆酸钠溶液,逆行注射到胰胆管内,把模型组及假手术组分成四个时间点,即3h、6h、12h、24h,观察各点的胰腺组织病理变化情况,通过western-blotting检测各组F-actin、RegⅢ的含量。结果:F-actin蛋白表达量在急性胰腺炎模型组中,呈降低趋势,和假手术组、空白组比较,差异具有统计学的意义(P<0.05)。RegⅢ蛋白表达量则明显升高在模型组中,与假手术组及空白组相比较,差异具有统计学意义的(P<0.05)。结论:大鼠急性胰腺炎的模型创建成功,用3%牛磺胆酸钠溶液,在胆胰管内逆行注入;F-actin蛋白表达量在急性胰腺炎模型组中,和空白组、假手术组比较,呈现减低的趋势;而RegⅢ表达量在模型组中,呈显著增高,和假手术组及空白组比较,在胰腺组织修复、再生方面RegⅢ起一定的作用。
Objective:To study the changes of F-actin and RegIII in pancreatic tissues induced by sodium taurocholate solution in acute pancreatitis for rats.Methods:Forty-five rats were randomly divided into acute pancreatitis model group,sham operation group and blank group with 15 rats in each group.Acute pancreatitis model was induced by injecting 3%sodium taurocholate solution into the pancreatic duct by retrograde cholangiopancreatography.The pathological changes of pancreas were observed at the 3h,6h,12h and 24h,and the contents of F-actin and RegIII in each group were detected by western-blotting.Results:The expression of F-actin protein in acute pancreatitis model group was lower than that in sham operation group and blank group(P<0.05).The expression of RegIII protein in model group was significantly increased than that in sham operation Group and the blank group,and the difference was statistically significant(P<0.05).Conclusion:The retrograde injection of 3%sodium taurocholate in pancreaticobiliary duct can induce the acute pancreatitis in rats.The expression of F-actin protein in the model group showed a decreasing trend compared with the blank group and the sham operation group.The expression of RegIII in the model group was significantly higher compared with blank Group,sham operation group.RegIII has a certain effect on pancreatic tissue repair and regeneration.
