摘要
本文分析了桑树SSR的实验过程:基因组DNA提取、引物筛选、PCR反应体系优化、引物退火温度选择、凝胶配制、电泳和凝胶染色、显色,并指出了这些步骤中需要注意的问题,为SSR技术在桑树研究中的应用提供基础。
In order to provide a foundation for the application of SSR(Simple Sequence Repeat)technology in mulberry research,this paper analyzes the experimental process of SSR used in mulberry:genomic DNA extraction,primer screening,optimization of SSR PCR reaction system,primer annealing temperature selection,gel preparation,electrophoresis,gel staining and color development,and points out the issues that should be paid attention to in these steps.
