虎粪便类固醇激素检测及影响因素的评估 被引量:3

摘要 粪便中类固醇激素的水平对动物的繁殖情况具有明确的指示意义。对于虎这一大型食肉动物,利用粪便激素水平来监测其繁殖状态是最为便捷的方式。但是,如果粪便中类固醇激素分布不均匀,不同性状的粪便和不同时间采集的粪便激素含量存在差异,那么在采样时就必须考虑这些因素。本研究用酶联免疫吸附检测法检测了雌性东北虎(Panthera tigris altaica)和华南虎(P.t.amoyensis)粪便中雌二醇、睾酮和孕酮这3种类固醇激素的水平,系统比较粪便表层、中层和中心的差异,不同采集时间所测得激素含量的差异,以及干燥和黏稠粪便中含量的差异。结果显示,睾酮、雌二醇和孕酮在表层、中层和中心各部分的含量无显著差异。在上午排出的粪便中孕酮的含量显著高于下午所排的粪便,而雌二醇和睾酮没有检测到显著差异。同一个粪团上较干燥部位和黏稠无定型的部位之间,3种激素的含量均没有显著差异。上述结果表明,采集虎的粪便时,可以不考虑粪便性状,除了分析孕酮之外,分析其他激素不需要考虑粪便排出的时间。在激素含量测定时,将整个粪便进行均质后再随机取样虽然不是必要的,但均质化仍有助于降低抽样引起的数据变差。 Fecal steroid hormones are indicators of reproductive status of animals.For large carnivores like tigers(Panthera tigris),it is convenient to use fecal hormone levels to monitor reproductive status.However,the effects of factors such as the quantity of fecal samples,location of sample material in fecal pellets(inner part or surface),and the elapsed time post-defecatation remain unclear.This study analyzed the content of metabolites of estradiol,testosterone and progesterone in feces of captive Amur tiger(P.t.altaica)and south China tiger(P.t.amoyensis)using enzyme-linked immunosorbent assay,and systematically compared the differences among fecal samples from the surface,middle and center of the pellets,between dry and sticky parts,and between feces defecated in the morning and afternoon.Samples from the surface,middle,and center of the pellets showed slight but not significant variation for all tested steroid hormones.The content of progesterone was significantly higher in feces defecated during morning as compared to afternoon,while estradiol and testosterone contents were similar.All three hormone contents were similar for dry and sticky part of fecal pellets.These results suggest that the effects of elapsed time of the feces should be considered when examining fecal progesterone levels in tigers.Completely mixing the samples from different parts of the pellets for all steroid hormones assay prior to subsequent analysis may not be necessary but homogenization is likely to reduce data variation raised from different sampling sites.
