摘要
目的研究组蛋白H3K79甲基化转移酶Dot1l在豚鼠耳蜗中的表达情况。方法将6只听力正常的豚鼠取材,运用免疫组织化学技术检测Dot1l在豚鼠耳蜗各转中的表达差异。结果 Dot1l表达于豚鼠耳蜗的血管纹、螺旋韧带、前庭膜、螺旋神经节、Corti器、螺旋缘、外螺旋沟细胞,在螺旋韧带的表达由底转向顶转减少(F=8.29,P=0.0091),在螺旋神经节中的表达由底转向顶转逐渐增多(F=6.22,P=0.0201),差异具有统计学意义。结论:豚鼠耳蜗中存在与ENa C表达相关的组蛋白H3K79甲基化转移酶Dot1l,其可能参与内淋巴循环。
Objective To study the expression of Disruptor of telomeric silencing 1-like(Dot1l)in cochleae of guinea pigs.Methods Six guinea pigs with normal hearing were sacrificed and expression of Dot1l in the cochlea was evaluated by immunohistochemistry.Rusults Dot1l was found in the stria vascularis,Reissner’s membrane,spiral ligament,organ of Corti,spiral limbus and spiral ganglion.From basal turn to apical turn,Dot1l expression in the spiral ligament significantly decreased(F=8.29,P=0.0091),while that in the spiral ganglion significantly increased(F=6.22,P=0.0201).Conclusion Dot1l,a protein that may be involved in regulation of ENaC expression,exists in the cochlea of guinea pigs.Dot1l in the cochlea may be involved in regulation of ENaC,and thus related to endolymph regulation.
