摘要
目的探讨枳壳多糖(Citrus aurantium L.polysaccharides-B,CALB)对^(60)Coγ-射线诱导小鼠辐射的保护作用。方法将BALB/c小鼠随机分为空白对照组、辐射模型组、CALB给药组(高、中、低剂量)、阳性对照组(黑木耳多糖,HP),连续灌胃给药30 d,末次给药3 h后,检测空白对照组及经^(60)Coγ-射线一次性全身照射(7 Gy)后,小鼠d 2、14的存活率。另检测空白对照组及经^(60)Coγ-射线一次性全身照射(3 Gy)后小鼠骨髓细胞DNA含量、骨髓细胞微核含量,小鼠脑组织、血清及肝脏中SOD、GSH-Px活性和MDA含量,以及脑组织中TCh E含量、小鼠脾、胸腺指数。结果 CALB各剂量组均能明显提高辐射小鼠的存活率,并提高小鼠骨髓细胞中DNA含量,降低骨髓细胞微核数。此外,CALB还可增加胸腺、脾指数,提高辐射小鼠血清、脑和肝组织中SOD、GSH-Px水平,同时降低MDA含量。结论 CALB对辐射损伤具有一定的保护作用,可用于枳壳的进一步开发与利用。
Aim To investigate the protective effect of Citrus aurantium L.polysaccharides-B(CALB)on radiation induced by 60 Coγ-ray in mice.Methods The BALB/c mice were randomly divided into blank control group,radiation model group,CALB administration group(high,medium and low dose),and positive control group(black fungus polysaccharide,HP).The mice were administered orally for 30 days.After the last administration for three hours,the survival rates on the 2nd day and the 14th day of the blank control group and the irradiated mice after the single radioactive irradiation(7 Gy)with 60 Coγ-ray were measured.In addition,DNA content and micronucleus of bone marrow cells,SOD,GSH-Px activities,MDA content in serum,liver and brain tissues in mice,TChE activity in brain tissues and spleen and thymus index of mice were detected after one-time whole body irradiation with 60 Coγ-ray(3 Gy).Results Each dose group of CALB could significantly improve the survival rate of irradiated mice,increase the DNA content of mouse bone marrow cells and reduce the number of micronuclei in bone marrow cells.In addition,CALB could also increase the thymus and spleen index and the levels of SOD and GSH-Px in serum,brain and liver tissues of mice,and reduce the content of MDA.Conclusion CALB has protective effect on radiation injury,which can be used for further development and utilization of Fructus aurantii.
