线纹海马活性多肽的体外抗氧化活性评价 被引量:2

摘要 近年来,随着人们对海马研究的深入,海马作为制备功能食品的原料受到越来越多的关注。通过研究洗脱流速、上样浓度及上样量对Sephadex G25树脂分离效果的影响,选择出最佳分离条件。利用Sephadex G25凝胶层析柱在最佳纯化条件下分离出不同分子量段的线纹海马混合肽,并对这些不同分子量组分体外抗氧化活性进行评价,以探究线纹海马多肽的抗氧化活性与分子量的关系。结果表明:0.4 mL/min的洗脱流速,0.10 mL的上样量,25 mg/mL的上样浓度作为Sephadex G25凝胶层析的最佳纯化条件。在此最佳纯化条件下分离出4个分子量段的多肽组分,通过测定各组分对DPPH自由基、超氧阴离子自由基和羟自由基清除率后发现,0.43 kDa^2.78 kDa的线纹海马多肽组分比其他分子量范围的组分有更好的抗氧化活性。 In recent years,as the research into the Hippocampus has been deepened,the Hippocampus has received increasing attention as the raw material for preparing functional foods.The relationship between the antioxidant activity and the molecular weight of the Hippocampus peptide was studied.First of all,the optimum separation conditions were selected by studying the effect of elution rate,loading concentration and loading on Sephadex G25 column separation.The mixed peptides with different molecular weight segments were separated under the best purification conditions using Sephadex G25 gel column.Then,the antioxidant activity of different molecular weight components were studied.The results showed that under the 0.4 mL/min elution rate,25 mg/mL loading concentration and 0.10 mL loading for Sephadex G25 column,0.43 kDa-2.78 kDa Hippocampal polypeptide component was higher than the other molecular weight,resulting in low molecular weight Hippocampus peptides with better antioxidant activity.
