miR-194在胃癌患者血液中的表达及临床意义 被引量:4

摘要 目的探讨miR-194在胃癌患者血液中的表达及临床意义。方法采集2015年2月至2016年7月120例胃癌患者和同期体检的40例健康对照者血液样本,实时RT-PCR法检测miR-194表达水平,并分析其表达水平与临床病理特征的关系。电化学发光法测定血清CA199、CA724、CEA含量;ELISA法检验miR-194靶基因BMI-1的表达水平,并分析miR-194表达水平与它们之间的相关性。结果胃癌患者血液中miR-194表达水平明显低于健康对照者(t=1.82,P=0.04);而肿瘤标志物水平CA199、CA724、CEA及靶基因BMI-1水平明显高于健康对照组(t=3.82,P=0.03;t=4.67,P=0.01;t=3.42,P=0.04;t=3.15,P=0.03)。miR-194的表达水平与患者的性别、年龄、肿瘤大小无关(P>0.05),与分化程度、TNM分期、淋巴转移有关(t=2.93,P=0.031;t=3.81,P=0.019;t=3.34,P=0.008)。miR-194表达水平与BMI-1水平呈负相关(r=-0.91,P<0.001),与肿瘤标记物CA199、CA724、CEA也分别呈负相关关系(r=-0.82,P=0.023;r=-0.94,P=0.007;r=-0.72,P=0.021)。结论 miR-194临床上可作为诊断胃癌的潜在生物标志物,可能通过调控BMI-1参与胃癌发生发展,与患者分化程度、TNM分期、淋巴转移密切相关。 Objective To investigate the expression and significance of miR-194 in the blood of patients with gastric cancer.Methods One hundred and twenty patients with gastric cancer who were treated in our hospital from February 2015 to July 2016 were enrolled in the study as observation group,moreover,the other 40 healthy subjects who received physical examination in our hospital at the same period were enrolled as control group.The expression levels of miR-194 were detected by RT-PCR.In addition the correlation between the expression levels of miR-194 and clinicopathological features were analyzed.Besides the serum levels of CA199,CA724 and CEA were detected by electrochemical luminescence assay.And the expression levels of BMI-1 were detected by ELISA.The correlation between the expression levels of miR-194 and the levels of BMI-1,CA199,CA724,CEA was analyzed,respectively.Results The expression levels of miR-194 in observation group were significantly lower than those in control group(P<0.05),however,the levels of CA199,CA724,CEA and target gene-BMI-1 were significantly higher than those in control group(P<0.05).Moreover the expression levels of miR-194 were not related with patient’s sex and age as well as tumor size(P>0.05),however,which were correlated with the differentiation degree of tumors,TNM staging,lymphatic metastasis(P<0.05 or P<0.01).Besides the expression levels of miR-194 were negatively correlated with those of BMI-1(r=-0.91,P<0.01),which were also negatively correlated with those of CA199,CA724,CEA(r=-0.82,P<0.05;r=-0.94,P<0.01;r=-0.72,P<0.05).Conclusion The miR-194 can be regarded as a potential biomarker in the diagnosis of gastric cancer,which may be involved in the pathogenesis and development of gastric cancer through regulating BMI-1,and it is closely related with the differentiation degree,TNM staging and lymphatic metastasis.
