摘要
【目的】提高粮食作物的营养品质就是改善人类自身的营养与健康。对水稻(Oryza sativa L.)等禾本科作物而言,种子蛋白质含量是一个极其重要的营养品质性状,解析种子蛋白质含量相关基因的功能具有十分重要的意义。【方法】以控制稻米蛋白质含量的主效QTL基因Os AAP6为研究对象,运用生物信息学方法,对其编码的蛋白理化性质、蛋白的结构及其功能进行分析。【结果】水稻OsAAP6基因cDNA序列全长为1401 bp,编码466个氨基酸,该蛋白为氨基酸透性酶,是氨基酸转运蛋白家族和APC超家族中的成员;序列比对结果显示,其它主要粮食作物(如小麦、大麦、玉米和高粱等)中有很多预测的基因与水稻OsAAP6基因存在较高的相似性;系统进化分析结果显示,在主要粮食作物中,水稻Os AAP6基因与高粱和玉米的进化关系最为密切,大麦次之,与小麦的亲缘关系较远。【结论】通过对水稻OsAAP6基因结构与功能的生物信息学分析,为主要粮食作物营养品质的遗传改良提供重要信息。
【Objective】Enhancing nutritional quality of food crops is a mean of improving human nutrition and health.Grain protein content(GPC)is an important nutritional quality trait existing in rice and many other crops,and it is of great significance to analyze the function of the genes related to the GPC.[M ethod]In this study,we used the bioinformatics methods to analyze the physical and chemical properties,the structure and function of the 0sAAP6 gene,which is one of the major QTL genes controlling GPC in rice.[R esu lt]T h e cDNA sequence of 0sAAP6 gene was 1401 bp,encoding a total of 466 amino acids,which was a membrane permeability enzyme of amino acids,belonging to the amino acid transporter family and the APC super gene family.The sequence alignment results unraveled that there was a high similarity between the 0sAAP6 gene and the predicted genes in the other major food crops,such as wheat,barley,maize and sorghum.The results of molecular phylogenetic analysis showed that the 0sAAP6 gene was most closely related to sorghum and m aize,followed by barley,but quite far from the phylogenetic location of wheat in the main grain crops.【Conclusion】The results of this study could provide important information for the genetic improvement of nutritional quality in rice and other staple crops.
