基于响应面法优化的苦荞多糖提取工艺及其抗疲劳作用研究 被引量:8

Optimization of Extraction Technology of Polysaccharides of Tartary Buckwheat Based on Response Surface Method and Its Anti-fatigue Effect

摘要 采用水提醇沉法提取苦荞多糖,利用响应面法中的中心组合设计,对提取工艺中的提取时间、提取温度和料液比进行考察.同时,建立小鼠负重游泳模型,给予小鼠不同剂量的苦荞多糖水溶液后,测定小鼠力竭游泳时间以及血乳酸、血尿素氮的浓度和肌糖原、肝糖原的含量.结果显示,利用响应面法得出的苦荞多糖最佳提取工艺条件为,液料比为16、提取温度为100℃、提取时间为120 min,多糖的得率在实测中达到了13.13%;服用苦荞多糖后可使小鼠的力竭游泳时间显著延长、肝糖原和肌糖原含量升高、而血乳酸和血尿素氮的浓度均有不同程度的降低.结果表明,苦荞多糖具有抗疲劳的作用. Based on the central composite design-response surface method,the extraction parameters of buckwheat polysaccharide including extraction time,extraction temperature,and liquid-solid ratio were primarily investigated.And then,crude buckwheat polysaccharides were obtained by ethanol precipitation method.A series of concentrations of polysaccharides were orally administrated to mice.By establishing the mice burden swimming model,the effect of polysaccharides on some indexes such as exhaustive swimming time,the concentrations of lactic acid and urea nitrogen in blood,and the amounts of glycogen in liver and muscle were evaluated after giving the mice Tartary buckwheat polysaccharides solution of different dose.The results showed that,the optimized extraction technology conditions of polysaccharides of tartary buckwheat based on response surface method were 16 folds of liquid-solid ratio,100 Tl of extraction temperature and 120 minutes of extraction time,with the highest extraction yield of 13.13%(w/w).After oral administration of buckwheat polysaccharides,the exhaustive swimming time of mice was significantly prolonged,and the glycogen amounts in liver and muscle increased.However,the lactic acid and urea nitrogen concentrations in blood decreased at different levels.It was concluded that the polysaccharides derived from Tartary buckwheat exhibited remarkably anti-fatigue activity.
