摘要
目的:研究季铵盐包裹溴化银纳米复合物(Ag Br/BHPVP)改性玻璃离子水门汀(GIC)的抗菌性能及细胞毒性。方法:分别制备含3%、4%、5%不同质量百分数的Ag Br/BHPVP-GIC试件作为实验组,同时制备不含Ag Br/BHPVP的富士Ⅱ型GIC试件作为对照组。采用琼脂扩散法检测各组试件对变异链球菌的抗菌能力,并利用活/死菌染色法及激光共聚焦显微镜(CLSM)观察各组试件表面的细菌活力;制备各组试件浸提液,并采用CCK-8法检测其对人牙髓细胞(h DPCs)的细胞毒性。结果:含不同质量百分数Ag Br/BHPVP的GIC各组对变异链球菌均具有一定的抗菌性,且其抑菌圈直径随着Ag Br/BHPVP添加量的增加而增大(P<0.05);各实验组的抑菌圈直径在1、7、14 d各时间点均大于对照组(P<0.05);同一组内相比,各组的抗菌性均未随时间而减弱(P>0.05)。CLSM观察结果显示,各改性GIC组试件表面粘附的活细菌数均明显少于对照组。细胞毒性实验结果显示,3%Ag Br/BHPVP细胞相对增殖率与对照组无统计学差异(P>0.05),Ag Br/BHPVP添加量为4%、5%时,细胞相对增殖率(%)为84.4±0.8、80.8±1,与对照组相比P<0.05。结论:在富士Ⅱ型玻璃离子中添加季铵盐包裹溴化银纳米复合物后,可增强其对变异链球菌的抗菌性能,添加量4%和5%时对h DPCs的细胞毒性为1级。
AIM:To investigate the antibacterial activity and cytotoxicity of glass ionomer cement(GIC)supplemented with a silver bromide/cationic polymer nanocomposite(AgBr/BHPVP).METHODS:AgBr/BHPVP were incorporated into the GIC at 3%,4%and 5%(w/w)respectively(test groups).A conventional GIC,FujiⅡcement was used as the control.The antibacterial activity of the materials on Streptococcus mutans was evaluated by agar-diffusion test on days 1,7 and 14 respectively.The biofilm staining of live and dead bacteria on the surface of the specimens was observed by confocal laser scanning microscope(CLSM).The cytotoxicity of the eluent of the samples to human dental pulp cells(hDPCs)was determined by CCK-8 assay.RESULTS:The highest inhibition zone of was found in the test goup of FujiⅡwith AgBr/BHPVP mass fraction of 5%.The inhibition zone increased with the addition of AgBr/BHPVP(P<0.05),and the antimicrobial activity did not decrease with time(P>0.05).CLSM indicated that the live bacteria on the specimens of the test groups were significantly fewer than those of control group.Cytotoxicity assay showed that there was no significant difference of cell proliferation between the goup of GIC-containing 3%(w/w)AgBr/BHPVP nanoparticles and the control group or the blank control group(P>0.05).But in the goup of GIC-containing 4%and 5%(w/w)AgBr/BHPVP nanoparticles the relative cell proliferation was(84.4±0.8)%and(80.8±1)%respectively(vs control,P<0.05).CONCLUSION:The addition of AgBr/BHPVP into FujiⅡGIC may increase antimicrobial properties over the conventional GIC alone for Streptococcus mutants.The cytotoxicity of GIC contiaining 4%and 5%AgBr/BHPVP to HDPCs is gradⅠ.
