大鼠疑核对内毒素炎症反应的影响及机制研究

摘要 为了探究大鼠疑核(NA)在炎症反应中的作用,选取大鼠腹腔注射内毒素(LPS)的方法来构建大鼠炎症模型,同时电损毁左、右侧的NA,采用免疫组织化学技术和酶联免疫(ELISA)实验技术,检测NA中即刻早期基因(Fos)和胶质纤维酸性蛋白(GFAP)的表达,以及血清中肿瘤坏死因子-α(TNF-α)和白细胞介素4(IL-4)的含量。结果表明:与对照组相比,LPS炎症组NA、迷走神经背核(DMV)、孤束核(NTS)中Fos和GFAP的表达具有显著的统计学意义,血清中TNF-α和IL-4的含量有明显的增大趋势;与假损毁左侧NA组相比,被损毁左侧NA组LPS炎症大鼠血清中IL-4以及TNF-α的水平具有显著统计学意义,同时大鼠NTS中的Fos的表达量明显增加;与假损毁组相比较,损毁右侧NA组血清中细胞因子TNF-α的含量没有显著变化,而IL-4含量显著增大,DMV和NTS中的Fos和GFAP表达也无明显变化;NA参与了LPS炎症反应,并通过左侧NA减少抗炎细胞因子的产生,增加促炎细胞因子的产生;右侧NA减少抗炎细胞因子产生,而对促炎细胞因子无调节作用,左右两侧NA在炎症反应中的作用可能有所差别。 To investigate the role of nucleus ambiguus in the inflammation,rats with left or right nucleus ambiguous(NA)electrolytic lesions or sham lesions were treated with intraperitoneal injection of lipopolysaccharide(LPS).Glial fibers acidic protein(GFAP)and Fos expression were tested by immunohistochemical staining,and serum levels of tumor necrosis factor-a(TNF-a)and interleukin-4(IL-4)were tested by enzyme linked immunosorbent assay(ELISA).The results show that in LPS group,the expressions of Fos and GFAP in NA,dorsal motor nucleus vagus(DMV),and nucleus of solitary tract(NTS)have significant statistical significance.The serum TNF-a,and IL-4level are significantly increased in comparison with the control group.Compared with the sham group(not has been electrical lesions),the serum TNF-a level in left NA lesioned rats is significantly decreased,while the level of IL-4is dramatically increased.The expression of Fos is significantly increased in left NA lesion rats.Compared with the sham group,IL-4levels are significantly increased,while there is no remarkable change in TNF-a level,Fos and GFAP expression in DMV and NTS in right NA lesioned rats.NA involves in LPS-induced inflammatory response.The left side of the NA may promote the production of proinflammatory factors and inhibit the production of anti-inflammatory factors.The right NA may inhibit anti-inflammatory factors production and has no effect on the production of proinflammatory factors.The left and right sides of NA may play different roles in the regulation of inflammatory response.
