摘要
以越橘品种‘科利尔’(Vaccinium vitis-idaea L.‘Koralle’)茎尖为试材,对影响茎尖离体培养的取材时期、外植体的消毒、培养基和激素配比等因素进行研究,旨在建立茎尖快繁体系。结果表明:休眠期水培促萌的嫩芽污染率较低,最利于茎尖诱导成活。采用正交试验设计筛选出最佳灭菌处理为75%酒精30 s+0.1%升汞4 min+2%次氯酸钠2 min,污染率和成活率分别为5.64%和84.15%;适于茎尖增殖培养的培养基和激素组合为WPM(改良)+1.0 mg·L-1ZT+0.1 mg·L-1GA3,增殖系数、苗高分别为4.85和2.74 cm。
In order to establish the shoottip rapid propagation system for blueberry,we studied the effect of sampling time,explant sterilization,culture medium and hormone combination on Vaccinium vitisidaea L.‘Koralle’shoottip.The tender bud,which promoted germination in water during dormant period,had lower pollution rate and higher survival rate.The better sterilization method by orthogonal design was directly surfacesterilized with 75%ethanol for 30s,0.1%HgCl2 for 4min and 2%NaClO for 2min,and the pollution rate and survival rate were 5.64%and 84.15%,respectively.The suitable multiplication culture medium and hormone combination for shoottip was:improved WPM+1.0mg·L-1 ZT+0.1mg·L-1 GA3,the multiplication coefficient and seedling height were 4.85 and 2.74cm,respectively.
