序贯高速离心消除脂血对常规生化项目检测干扰的效果评价 被引量:9

摘要 目的:评价序贯高速离心消除脂血对常规生化项目检测干扰的效果。方法 :通过将脂血血浆于试管内直立过夜获得上层脂质,与澄清血浆(原始血浆)按一定比例混合制造脂血模型,脂血模型以序贯高速离心后测20项生化指标,并与原始血浆的生化指标结果比较。结果:6.55 mmol/L三酰甘油(triglyceride,TG)脂血有Cl^-、Mg^(2+)、P^(3-)、谷丙转氨酶(alanine aminotransferase,ALT)、总蛋白(total protein,TP)、白蛋白(albumin,ALB)、总胆红素(total bilirubin,TBIL)、谷氨酰转肽酶(glutamyl transpeptidase,GGT)、碱性磷酸酶(alkaline phosphatase,ALP)、乳酸脱氢酶(lactate dehydrogenase,LDH)、淀粉酶(amylase,AMY)、尿素氮(blood urea nitrogen,BUN)的偏倚超过中国临床生物化学常规检验项目分析质量标准,10.48 mmol/L TG脂血除上述指标外,还有肌酸激酶(creatine kinase,CK)、谷草转氨酶(aspartate transaminase,AST)的偏倚超过国家规定的标准;序贯高速离心处理后,Mg^(2+)、P^(3-)、ALT、AST、ALB、ALP、LDH、AMY偏倚不大于国家规定的标准,Cl^-、TP、TBIL、GGT、CK、BUN大于国家规定的标准。结论 :本实验室的条件下序贯高速离心处理能有效去除脂质对Mg^(2+)、P^(3-)、ALT、AST、ALB、ALP、LDH、AMY检测的影响。 Objective:The aim of this study was to evaluate the effect of eliminating lipemia interference in routine biochemical tests by using consecutive high speed centrifugation.Methods:Lipid is harvested through filling the lipemia to test tube and let standing overnight at4℃.Aliquots of the clarified plasma(native plasma)were spiked with lipid to make lipemia model.The lipemia was removed from the spiked samples using consecutive high speed centrifugation.Afterwards,the biochemical parameters were measured.The parameters analysed were compared to the parameters of native plasma.Results:The bias of the spiked samples exceeded the quality specification of china at6.55mmol/L triglyceride(TG)for Cl-,Mg2+,P3-,alanine aminotransferase(ALT),total protein(TP),albumin(ALB),total bilirubin(TBIL),glutamyl transpeptidase(GGT),alkaline phosphatase(ALP),lactate dehydrogenase(LDH),amylase(AMY),blood urea nitrogen(BUN);and for all previously mentioned parameters including creatine kinase(CK),aspartate transaminase(AST)at concentration of10.48mmol/L TG.The bias of the spiked samples met the quality specification of china for Mg2+,P3-,ALT,AST,ALB,ALP,LDH,AMY after using consecutive high speed centrifugation.Conclusions:Consecutive high speed centrifugation is suitable for lipemia removal from samples designated for Mg2+,P3-,ALT,AST,ALB,ALP,LDH,AMY our laboratory.
