HPLC法检测牛奶中喹诺酮类药物残留色谱条件的优化 被引量:1

摘要 通过对GB/29692-2013方法一中流动相组成比例、流动相pH值、色谱柱规格三个色谱条件的研究,发现在满足药物色谱峰分离度>3.0的条件下,应用CAPCELL PAK C18(250 mm×4.6 mm,5μm)色谱柱,流动相磷酸三乙胺溶液与乙腈的比例为80:20时药物保留时间短;色谱柱规格为C18(100 mm×4.6 mm,2.6μm)的情况下,药物保留时间短;pH值在2.0~4.0的范围内对药物保留时间影响不显著。因此,按照国标中方法一进行检测时,流动相中乙腈组分的比例越大,色谱柱的长度越短,填料粒径越小检测效率越高。 Through the optimization of GB/29692-2013method in a mobile phase ratio,pH value of mobile phase,specifications and models of chromatographic column,found in the chromatographic peak resolution of>3.0,the retention time of quinolonesis short when using chromatographic column CAPCELL PAK C18(250mm×4.6mm,5μm)with phosphoric acid and triethylamine solution to acetonitrile80:20ratio.The retention time of quinolonesis short when using chromatographic column C18(100mm×4.6mm,2.6μm),the retention time of quinolonesis is not significantly affected when the pH value in the range of2.0~4.0.Therefore,according to the method in the national standard,the larger proportion of acetonitrile in the mobile phase,the shorter length of the chromatographic column and the smaller size of the chromatographic column packing,the higher of the detection efficiency.
