摘要
肿瘤坏死因子-α(TNF-α)是一种具有多种生物学活性的细胞因子,其过量表达与多种疾病密切相关。TNF-α抑制剂可以抑制TNF-α与受体TNFR1之间的结合,破坏二者之间的信号传导,抑制TNF-α过度表达所引起的一系列疾病。已开发的抑制剂主要包括合成抗体、合成小分子、天然产物、金属和多肽,本文将对一系列抑制剂作一综述。
Tumor necrosis factor-α(TNF-α)is a cytokine with a variety of biological activity.Its overexpression is closely related to various diseases.TNF-αinhibitors can inhibit the binding of TNF-αto the receptor TNFR1,destroy the signal transduction between them,and inhibit a series of diseases caused by TNF-αover-expression.Developed inhibitors include synthetic antibodies,synthetic small molecules,natural products,metals and peptides,a series of inhibitors will be reviewed in this paper.
