摘要
铁皮石斛作为一种重要的抗癌抑癌药物,在全民养生的今天越来越多的成为了人们消费的热点。然而,由于野生的铁皮石斛非常稀少而且非常昂贵,不适合大众消费。所以建立一套适合工业化生产的组培体系成为了发展的趋势。以幼嫩的铁皮石斛带节茎段为原材料,1/2MS作为培养基,探究了不同激素浓度及其他因素对铁皮石斛快繁和组培的影响。再以组培出的幼苗为原材料,研究了温度、水质、培养基质及浇水方式等对铁皮石斛炼苗的影响。结果表明,铁皮石斛快繁的最佳激素浓度为6-BA,1.0 mg/L和NAA,1.0 mg/L,及1%的活性炭,诱导率为12.5%;组培的最佳激素浓度为6-BA,1.5 mg/L及NAA,2.0 mg/L或6-BA,0.5 mg/L,NAA,0.5 mg/L和2,4-D,1.5 mg/L,诱导率都在80%以上。铁皮石斛的最优炼苗条件为25℃,光照过的自来水浇灌;最适合的培养基质为水苔+直径约为0.5~1.0cm的树皮。
As an important medicine for anticancer,Dendrobium officinale Kimura et Migo becomes a hot topic during thehealth-preserving society.Wild Dendrobium officinale is very rare and very expensive and not suitable for publicconsumption.So the establishment of the tissue culture system suitable for industrial production has become the tendency.We used its stalk as material and1/2MS as basal culture medium,investigated the effects of different hormone concentrationand other factors on the tissue culture of D.Officinale,and then used these plants cultivated as material to explore the effectsof temperature,water quality,culture medium and watering method on the seedling of Dendrobium officinale.The resultsshowed that the best medium of rapid multiplication was basal culture medium+1.0mg/L6-BA+1.0mg/LNAA+1%PAC,and it’s inductivity was12.5%.The best medium of tissue culture was basal culture medium+1.5mg/L6-BA+2.0mg/LNAA or0.5mg/L6-BA+0.5mg/L NAA or0.5mg/L NAA+1.5mg/L2,4-D,and their inductivities were all over80%.Thebest conditions to hardening-seedlings was25℃+lighted tap wate,the most suitable culture medium was sphagna+0.5~1.0cm barks in diameter.
