摘要 本文报告家兔主动脉平滑肌细胞(简称SMC)贴块和传代培养方法及对SMC形态学组织化学的初步观察.贴块培养5天后,SMC开始从组织块边缘长出并向外迁移,3周后SMC围绕组织块形成一细胞相互平行排列的环形致密层。传代的SMC贴壁后先汇聚成束状,然后呈现典型的“峰谷”样生长。SMC呈带状或梭形,有一卵圆形或腊肠状的核,核仁明显。SMC的线粒体数量少位于核周细胞质中。传代的SMC胞质中有较多的丝样结构。SMC的琥珀酸脱氢酶(SDH)、ATP酶(ATPase,PH9.2)反应阳性。比较而言,传代SMC其SDH、ATPase活性及线粒体数量都较贴块培养的SMC明显。在两种方法的培养过程中均未见到SMC的自主性收缩。 The article reports the culture methods of substrate-attached explants and Subcultures of aorta smooth muscle cells (SMCs) of rabbit and the preliminary morphological and histochemical observation for these cells. SMCs in explants attached to the substrate began to grow along the substrate and migrate away from the explants. After 3 week, a continuous circular sheet of cells formed into oriented Parallel bundles around the explant. In subculture, the dense aggregates of SMCs grew into bundles and showed characteristic hill and valley morphology. The SMCs have ribbon, spindle or astroid shape with one oval or sausage-shaped nucleus containing one or three nucleoli. SMCs have few mitochondria which are usually found in the perinuclear region. In subcultures, the SMCs appear to have many long. straight filament-like structures scattering along the cell longitudinal axis in the cytoplasm. SMCs show positive reaction of succinate dehydrogenase (SDH) and adenosine triphospalase (ATPase PH=9.2)and these enzyme reaction products scattered diffusely throughat the cytoplasm. SMCs in subculture have more mitochondria and greater SDH and ATPase activities than SMCs from explants do. It displays that the mitochondria and enzyme activity of the subcultured cells have a tendency of increase as culture goes on. spontaneous contraction has never been found in SMCs of substrate-attached explants and subcultures.
