摘要 目的探讨神经节苷脂(GLS)对大鼠脑缺血后功能恢复的影响及其可能机制。方法用线 栓法制备局灶性脑缺血模型,缺血后30min及3h,腹腔注射GLS(30mg/kg),以后1次/d, 共21d。用Bederson评分来评价神经功能缺失,并利用神经细胞培养观察GLS治疗后脑组织提 取液(BTE)中神经营养因子(NTFS)的活性变化。结果GLS可显著促进鼠脑缺血后神经功能 缺失的恢复。GLS治疗组BTE维持培养神经元存活的总数和长出神经突起超过100μm的神经元 数目较对照组明显增加(P< 0.05)。结论GLS可能通过增强BTE中NTFS的作用促进脑缺血后 神经功能的恢复。 Objective To study if ganglioside (GLS) may promote the recovery the ne urological deficits after cerebral ischemia in rats and the me chanism about it. Method Focal cerebral ischemia was made by a 3-hour occlusion of the right midd le cerebral artery(MCA)using nylon monofi1ament in rats and was fo1lowed by repe rfusion.GLS was intraperitoneally administered 30 minutes and 3 hours after isch emia and daily during the observation period. Bederson’s method was used to eval uate the neurological deficits. The activity of the neurotrophic factors (NTFs) in the periischemic brain tissue extract (BTE) and the effect of GLS on the acti vity were observed in cultured neuronsfrom the newbom rat cortex and basal gangl ia.Result GLS improved the neurologicalfUnction recovery after cerebral ischemia . The BTE of GLS treatment groap supportedneuronal survival and promoted neurite outgrowth about l00 μm. The number of neuronswas significantly higher in the p resence of the BTE from treatment grouP as compared with the BTE from control gr oup (P&lt; 0.05).Conclusion It indicated that GLS improved the recovery of the neur ological deficits by potentiating the activity of neuro trophic factors.
