摘要
目的 了解广西肝癌高发区儿童氧化性DNA损伤情况。方法 采用高效液相色谱 电化学方法检测广西扶绥县肝癌户儿童 2 1例及对照南宁市儿童 6 3例尿液 8 羟基脱氧鸟苷 ( 8 OH dG)水平。结果 扶缓 南宁两地儿童间尿 8 OH dG水平无显著性差异 (P =0 .98)。本组 8 OH dG[( 5 2 6± 0 4 1 )ng/mg肌酐 ,n =84 ],高于同类研究的检测值 [( 4 6 2± 0 0 91 )ng/mg肌酐 ,n =32 ];并与年龄 (r =- 0 .4 5,P <0 0 0 1 )、体质指数 (r =- 0 .2 7,P =0 .0 1 )显著负相关。多元分析提示年幼者(P =0 .0 0 0 1 )和体瘦者 (P =0 .0 3) 8 OH dG水平高 ,并且 8 OH dG与ALT的偏相关性检验 ,P =0 .0 71为临界值。结论 这是基于肝癌高发区高危人群的尿 8 OH dG分析的首次报道 ;提示引起该人群 8 OH dG升高的因素较为复杂 ;并且对青少年还应考虑生长发育因素的影响。
Objectives To explore oxidative DNA damage in children from liver cancer high risk areas in Guangxi Autonomous Region.Methods The urinary level of 8 hydroxydeoxyguanosine (8 OH dG) in 21 cases of children from hepatocellular carcinoma families in Fusui County and 63 controls from Nanning City were measured by high pressure liquid chromatography electrochemical detector(HPLC ECD)system.Results The mean 8 OH dG level (ng/mg of creatinine)in Fusui group was 5.26±0.41 SE which was higher than that in previous preport (4.62±0.091 SE) by similar method.No significant difference of 8 OH dG level between Fusui group and Nanning group was observed ( P =0.98).The 8 OH dG level was significantly negatively correlated with age ( r =-0.45, P < 0.001 ) and body mass index (BMI)( r =-0.27, P =0.01).In a multi regression analysis the 8 OH dG level in the juniors and the children with low BMI( P =0.03)was significantly high,and correlated with alanine aminotransferase (ALT)at critical level ( P = 0.071 ).Conclusion This study is the first report on oxidative DNA damage based on analysis of urinary 8 OH dG among the children from liver cancer high risk population.The data suggest that various factors might impact the 8 OH dG level among the unique population.An extensive study combined with other parameters should be necessary for further elucidation of oxidative DNA damage.
