摘要 目的 探讨金黄色葡萄球菌肠毒素B(SEB)对小鼠树突状细胞 (DC)表面I-Eκ 分子表达的影响。方法 密度梯度离心获得小鼠脾单个核细胞 ,Panning法纯化非贴壁细胞得到DC ;免疫组化SABC法检测DC纯度 ;流式细胞术检测SEB刺激后DC表面I -Eκ 分子的表达。结果 DC纯度为 70 %~ 80 %。I -Eκ 的表达在 10 0ng/mlSEB刺激 8h后达到高峰。结论 SEB可显著提高DC表面I -Eκ 分子的表达 ,该刺激作用有一最佳量效和时效关系。 Objective To investigate the effect of Staphylococcal enterotoxin B (SEB) on the expression of I-E κon the mice splenic Dendritic cell (DC).Methods The mice splenic mononuclar cells were separated by centrifugal in density gradient (Ficoll-Hypaque), then DCs were isolated by panning, and the purity of DCs was identified by SABC; After stimulated by SEB at different time and different dose, the I-E κ expression on DC was analyzed by Flow Cytometer.Results The purity of DC was 70%~80%, and the expression of 100ng/ml SEB.I-E κ molecule reached peak at 8 hour treated by Conclusions SEB can significantly enhance the expression of I-E κ molecule on the DC surface, and there is a certain relation between the effect and stimulating time and dose.
