摘要
目的观察金黄色葡萄球菌肠毒素B(staphylococcal enterotoxinB,SEB)对体外自然杀伤细胞(NK)细胞抑制T24膀胱癌细胞增殖及致细胞凋亡作用的影响,探讨SEB尾静脉注射对小鼠T24细胞皮下移植性膀胱肿瘤生长的作用。方法体外细胞实验采用补体裂解法分离收集人血NK细胞,用不同浓度的SEB(10^-5、10^-4和10^-3g/L)与NK细胞作用12h,然后取NK细胞加入T24细胞培养皿中,于12h、24h和48h后测定T24细胞的细胞活力(MTT法)和细胞凋亡率(流式细胞术)。动物实验建立T24细胞小鼠皮下移植膀胱肿瘤模型,30只小鼠随机分为生理盐水组、环磷酰胺组和SEB组(10、50、250μg/kg),检测各组肿瘤重量、抑瘤率,测定血浆和肿瘤TNF—α和IFN-γ水平(ELISA法)。结果NK细胞经过SEB处理后,T24细胞活力显著降低,凋亡率升高,具有显著的时间和剂量效应。SEB组肿瘤重量明显低于生理盐水组,抑瘤率接近环磷酰胺组,具有时间和剂量效应。血浆和肿瘤TNF—α和IFN-γ水平均高于其余两组。结论SEB可通过NK细胞作用及相关细胞因子等显著抑制T24细胞的体外活力及在体生长。
Objective To investigate the effect of staphylococcal enterotoxin B(SEB) on the proliferation and apoptosis of bladder carcinoma cell line T24 which was induced by NK cells, and to explore the effect of SEB through caudal intravenous injection on the growth of T24 subcutaneous tumor of mice. Methods Complement lysis method was used to collect human blood NK cells which were then treated with different concentrations of SEB (10-5 g/L, 10-4 g/L and 10-3g/L)for 12 h. Afterwards, the treated NK cells were co - cultured with T24 cells for 12,24 or 48 h. Viability and apoptosis rate of T24 cells were assayed with MTTand FCM, respectively. Subcutaneous tumor model of T2 cells was established in mice. A total of 30 mice were randomly divided into saline group, cyclophosphamide group and SEB groups (10,50, 250 μg/kg respectively). Tumor weight and inhibition rate were determined. TNF - α and IFN - γ, levels of the tumor tissue and plasma were detected with ELISA. Results After co - culture with SEB - treated NK cells, the vitality of T4 cells was reduced significantly;the apoptosis rate was significant increased time - and dose - dependently ; tumor weight was much lower than that in the saline group. Inhibition rate in the SEB groups was close to that in the cyclophosphamide group. TNF - α and IFN - γ, levels in the plasma and tumor tissue were much higher than those in the saline group and the cyclophosphamide group. Conclusion SEB can significantly inhibit the proliferation of T24 cells in vitro and in vivo when combined with NK cells.
