An Illumina-Based Analysis of the Diversity of Endophytic Bacteria from Different Mulberry Genotypes

摘要 为了分析不同抗病性桑树品种内生细菌多样性,利用基于Illumina MiSeq的高通量测序技术,从桑椹菌核病不同抗性品种(抗性品种:长果桑与川桑7637;易感品种:新伦教与红果2号)中共获得有效序列92282条,将高于97%相似度的划分为一个操作分类单元(OTU),优化后得到5858个OTUs。内生细菌种群分析结果表明,变形菌门(Proteobacteria,36.10%)、厚壁菌门(Firmicutes,14.51%)、酸杆菌门(Acidobacteria,13.38%)、拟杆菌门(Bacteroidetes,11.85%)以及放线菌门(Actinobacteria,6.35%)为各品种果桑的共有优势菌门,假单胞菌属(Pseudomonas,8.01%)为其共有优势菌属,但这些优势菌群在不同品种桑树中所占的比例均有差异。内生细菌多样性分析结果表明,抗性品种川桑7637的内生细菌多样性最高,长果桑次之,而易感品种新伦教与红果2号则相对较低。研究结果显示,桑树茎内具有丰富的内生细菌资源,抗性品种的内生细菌多样性高于易感品种。 In order to understand the relationship between the disease resistance of mulberry and the characteristics of endophytic bacterial community, the diversity of endophytic bacteria from different disease-resistant mulberry genotypes was researched with the Illumina MiSeq high-throughput sequencing technology, and 92282 effective sequences were obtained from the mulberry sclerotiniose-resistant varieties Changguo Sang and Chuan Sang No.7637 and the susceptible varieties Xin Lunjiao and Hong Guo No.2. A total of 5858 operational taxonomic units (OTUs) were collected at the 3% cutoff level after the sequences were optimized. The taxonomic analysis of endophytic bacteria showed that Proteobacteria (36.10%), Firmicutes (14.51%), Acidobacteria (13.38%), Bacteroidetes (11.85%), and Actinobacteria (6.35%) were the common dominant bacteria of all mulberry cultivars at the phylum level, and Pseudomonas was a prevalent genus, but the proportion of these bacteria in different mulberry samples were different. The diversity analysis results showed that the bacterial community diversity of the resistant variety Chuan Sang No.7637 was the highest, followed by Changguo Sang, while that of the susceptible varieties Xin Lunjiao and Hong Guo No.2 was relatively low. The above results showed that the mulberry stem harbors great amount of endophytic bacteria, and the endophytic bacterial diversity of resistant varieties is higher than that of susceptible varieties.
