MEBO对SD大鼠深Ⅱ度烫伤创面微循环影响的实验研究 被引量:2

摘要 目的探讨湿润烧伤膏(moist exposed burn ointment,MEBO)对SD大鼠深Ⅱ度烫伤创面微循环的作用机制。方法选取65只SD雄性大鼠,每只大鼠背部建立2个烫伤创面并随机分为MEBO组与对照组,其中MEBO组创面采用MEBO换药治疗,对照组不做任何处理,观察两组创面组织中内皮素-1 (ET-1)、一氧化氮合成酶(NOS)、热休克蛋白70 (HSP70)、CD34及髓过氧化物酶(MPO)的变化情况。结果 (1)大体观,MEBO组创面始终保持湿润状态,新生肉芽组织及上皮组织逐渐生长,至烫伤后第16天创面基本愈合;对照组创面始终干燥坚硬,至烫伤后第16天焦痂完全脱落,大部分创面愈合,创面明显缩小。(2)两组创面愈合率对比,其中烫伤后第4、7、19天,P均> 0. 05,差异无统计学意义;烫伤后第10、13、16天,P均<0. 05,差异具有统计学意义。(3)烫伤后1、6、12、24、48、72、96、120、144 h两组创面组织中ET-1、NOS及HSP70表达水平对比,P均> 0. 05,差异无统计学意义。(4)烫伤后第1、4、7、10、13、16天两组创面组织中MPO表达水平对比,P均> 0. 05,差异无统计学意义。(5)两组创面组织中CD34表达水平对比,其中烫伤后第1、4、10、13、16天对比,P均> 0. 05,差异无统计学意义;烫伤后第7天对比,P <0. 05,差异具有统计学意义。结论MEBO可促进SD大鼠深Ⅱ度烫伤创面的愈合,但与微循环相关介质ET-1、NOS、CD34等并无明显相关性,MEBO促进创面愈合的作用机制仍需进一步深入研究探讨。 Objective To investigate the action mechanism of MEBO( Moist Exposed Burn Ointment) on microcirculation of deep second scald wounds in SD rats. Methods Sixty-five SD male rats were selected as subjects. Make two scald wounds at the back of each rat and randomly divide the wounds into MEBO group and control group. The wounds in the MEBO group was treated with the dressing change of MEBO while the wounds in the control group were left alone and not treated at all. The changes of endothelin-1( ET-1),nitric oxide synthase( NOS),heat shock protein 70( HS70),CD34 and myeloperoxidase were observed in wounds of the two groups. Results( 1) Based on general observation,wounds inthe MEBO group can maintain moist all the time,new granulation tissues and epithelial tissues gradually grew well,and the wounds were basically healed on day 16 after scalding; while wounds in the control group were always dry and hard,on day16 after scalding,eschar all fell off,most wounds were healed and the residual wounds were significantly decreased in size.( 2) The wound healing rates were compared between the two groups on day 4,7 and 19 after scalding and the results showed no statistically significant difference( all P〈0. 05),while the comparison on day 10,13 and 16 showed statistically significant difference( all P〈0. 05).( 3) The expression levels of ET-1,NOS and HSP70 in wounds were compared between the two groups at 1 h,6 h,12 h,24 h,48 h,72 h,96 h,120 h and 144 h after scalding and no statistically significant difference was observed( all P〈0. 05).( 4) The expression levels of MPO in wounds were compared between the two groups on day 1,4,7,10,13 and 16 after scalding and no statistically significant difference was observed( all P〈0. 05).( 5) The expression levels of CD34 in wounds were compared between two groups on day 1,4,10,13 and 16 after scalding and the results showed no statistically significant difference( all P〈0. 05),while the comparison on day 7 after scalding
